Summary: ABC transporters are expressed in skin cells to protect against harmful xenobiotics and ensure skin homeostasis. Specific ABC proteins, such as MDR1/3, TAP1/2, CFTR, SUR1/2, and BCRP, play important roles in the skin. UV radiation increases the activity of ABC transporters, while oxidative stress affects mitochondrial SURs. However, there is limited experimental research on the physiological and pathophysiological roles of ABC transporters in skin cells.

Progress in understanding their role in health and disease, recognized transporter substrates, post-translational regulation, microRNA involvement, and structural elements important for transport activity are discussed. Additionally, the impact of lipids such as cholesterol on ABCG2 transport function and a potential model of cholesterol binding sites for ABCG2 are described.
