Atheroprotective effect of dietary walnut intake in ApoE-deficient mice: Involvement of lipids and coagulation factors

Introduction: Consumption of n-3 polyunsaturated fatty acids (PUFA) and antioxidant polyphenols is considered to decline the risk of cardiovascular disease. Materials and Methods: To provide an explanation for this cardioprotective effect, we performed an intervention study with proatherogenic Apoe(-/-) mice which were fed during eight weeks with a high fat diet supplemented with either walnuts (rich in n-3 PUFA and antioxidant compounds), walnut oil (with n-3 PUFA only) or sunflower oil as a control (12 mice per group). Results: Feeding walnuts, but not walnut oil, caused a 55% reduction in atherosclerotic plaque development in the aortic arch in comparison to the control diet. This was associated with reduced staining of plaques for CD36, a scavenger receptor expressed by macrophages. Feeding mice with walnuts also lowered plasma levels of triglycerides, cholesterol and prothrombin with 36%, 23% and 21 %, respectively, compared to control diet. In addition, accumulation of lipids in the liver was decreased, while plasma antioxidant capacity was increased. On the other hand, feeding mice with walnut oil did not provoke significant changes in these parameters in comparison to the control diet. Platelet activation and thrombus formation under flow remained unchanged with either diet. Conclusions: In Apoe(-/-) mice on high fat diet, intake of dietary walnut (but not walnut oil) beneficially influences lipid metabolism and atherosclerotic plaque development, with no more than limited effects on platelet and coagulation function. This method has promising applications in biomedical and clinical research.

Inhibition of Src but not Syk causes weak reversal of GPVI-mediated platelet aggregation measured by light transmission aggregometry

Summary: Src tyrosine kinases and spleen tyrosine kinase (Syk) play a role in sustained platelet aggregation under minimal shear following activation of glycoprotein VI (GPVI) and C-type lectin-like receptor-2 (CLEC-2). Aggregation and tyrosine phosphorylation were monitored by light transmission aggregometry (LTA) and fluorescence-labeled antibody. Src contributes to sustained aggregation through a Syk-independent pathway that inhibits outside-in signaling from glycoprotein IIb-IIia (GPIIIb-IIIa) to the cytoskeleton.

Gut enterochromaffin cells drive visceral pain and anxiety

Summary: Gastrointestinal discomfort is a common symptom of gut disorders and chronic visceral pain, especially in patients with irritable bowel syndrome. Enterochromaffin (EC) cells, which are rare serotonergic neuroendocrine cells in the gut epithelium, play a crucial role in detecting and transmitting noxious stimuli to nerve endings. The activation and dysfunction of EC cells are associated with hypersensitivity to gut distension and the inflammatory effects of certain fatty acids. Additionally, prolonged EC cell activation leads to persistent visceral hypersensitivity and anxiety-like behaviors. Furthermore, there are notable sex differences in the involvement of the EC cell-mucosal afferent circuit.

Unraveling the Antioxidant Activity of 2R,3R-dihydroquercetin

Summary: It has been found that 2R,3R-DHQ does not convert into quercetin in an oxidative environment, but forms an unstable oxidation product. The addition of glutathione leads to the formation of a 2R,3R-DHQ-GSH adduct, indicating oxidation of the B-ring. Ascorbate can reduce the quinone back to 2R,3R-DHQ. This suggests that the redox-modulating activity of 2R,3R-DHQ quinone resides in its B-ring.

Summary: This study investigated the roles of different extracellular matrix proteoglycans in platelet responses and thrombus formation. It was found that one proteoglycan (perlecan) enhanced platelet activation and promoted thrombus formation, but also reduced platelet adhesion under flow. This suggests that proteoglycans play an important role in platelet activation and thrombus formation.

Cross-Sectional Evaluation of Food Items Preferred by Adolescents under the Influence of Television Advertisements

Summary: This study examined the food and beverages purchased by Turkish adolescents under the influence of television advertisements and evaluated the products using the WHO nutrient profiling model. Results showed that unhealthy foods, such as cakes, confectionery, savory snacks, and soft drinks, were heavily advertised and purchased by the adolescents. Only a small proportion of the advertised products were deemed healthy according to the WHO nutrient profile model.

Adhesive properties of plasma-circulating and platelet-derived microvesicles from healthy individuals

Summary: In this study, we investigated the adhesive properties of circulating plasma-derived and platelet-derived microvesicles (MVs) and their potential role in thrombus growth. The results showed that both types of MVs weakly bound fibrinogen but did not stably adhere to immobilized fibrinogen. Moreover, the presence of these MVs did not significantly affect thrombus formation. Therefore, MVs present in the plasma of healthy individuals are unlikely to directly contribute to thrombus formation under arterial flow conditions.

Thrombotic triad in microgravity

Summary: Thrombotic disease may be underdiagnosed in microgravity conditions, and the underlying factors are still poorly understood. A case of internal jugular vein thrombosis was recently diagnosed in a low-risk female astronaut, and six out of ten additional crew members showed risk factors for jugular venous flow. Observations in space and studies on Earth suggest that microgravity affects blood flow, coagulation, and vascular function.

Driver gene KRAS aggravates cancer-associated stroke outcomes

Summary: The incidence of cancer-associated stroke has increased with the prolonged survival times of cancer patients. In this study, the researchers focused on the driver gene KRAS and found that KRAS mutations exacerbated neurological deficits after stroke and promoted tumor growth. They also identified the upregulation of pro-inflammatory responses and the promotion of STAT3 phosphorylation in mice with KRAS mutations. Additionally, the researchers found that colorectal cancer patients with stroke onset within 3 months after cancer diagnosis had a worse prognosis.

Safety of fixed-dose heparin titration boluses in underweight patients
