Evaluation of Antioxidant and DNA Damage Protection Activity of the Hydroalcoholic Extract of Desmostachya bipinnata L. Stapf

Desmostachya bipinnata Stapf (Poaceae/Gramineae) is an official drug of ayurvedic pharmacopoeia. Various parts of this plant were used extensively in traditional and folklore medicine to cure various human ailments. The present study was aimed to evaluate the antioxidant and DNA damage protection activity of hydroalcoholic extract of Desmostachya bipinnata both in vitro and in vivo, to provide scientific basis for traditional usage of this plant. The extract showed significant antioxidant activity in a dose-dependent manner with an IC50 value of 264.18 +/- 3.47 mu g/mL in H2O2 scavenging assay and prevented the oxidative damage to DNA in presence of DNA damaging agent (Fenton's reagent) at a concentration of 50 mu g/mL. Also, the presence of extract protected yeast cells in a dose-dependent manner against DNA damaging agent (Hydroxyurea) in spot assay. Moreover, the presence of extract exhibited significant antioxidant activity in vivo by protecting yeast cells against oxidative stressing agent (H2O2). Altogether, the results of current study revealed that Desmostachya bipinnata is a potential source of antioxidants and lends pharmacological credence to the ethnomedical use of this plant in traditional system of medicine, justifying its therapeutic application for free-radical-induced diseases.

This review provides an overview of the significant impact of selenium in different cancers, discussing its anti-cancer mechanisms and signaling pathways.

Summary: The organization of eukaryotic genome in the nucleus is complex and dynamic, and its morphology and size have significant impact on nuclear mechanics, chromatin organization, gene expression, cell functionality, and disease development. Abnormal nuclear envelope morphologies are implicated in human disorders. However, our knowledge about the molecular mechanisms regulating nuclear morphology and cell functionality during health and illness is limited. This review discusses the essential components governing nuclear organization and the functional consequences associated with nuclear morphometric aberrations, as well as recent developments with diagnostic and therapeutic implications for nuclear morphology.

Summary: This study characterized the anti-proliferative and apoptotic effects of a novel small molecule DJ4 in non-small cell lung cancer and triple-negative breast cancer cells. Structural modifications led to the discovery of four active analogs with enhanced ROCK/MRCK inhibitory potency. These active analogs showed anti-proliferative and anti-migratory effects in various cancer cell lines.

ABHD11-AS1: An Emerging Long Non-Coding RNA (lncRNA) with Clinical Significance in Human Malignancies

Summary: The aberrant expression of lncRNA ABHD11-AS1 has been linked to the development and progression of various cancers, with its high expression observed in gastric, lung, breast, colorectal, thyroid, pancreas, ovary, endometrium, cervix, and bladder cancers. It has clinical significance in prognosis, diagnosis, prediction of cancer progression stage, and treatment response. ABHD11-AS1 exerts oncogenic effects by altering various mechanisms and has prognostic potential in certain cancers. However, further research is needed to elucidate the specific role of ABHD11-AS1 in tumor development, prognosis, progression, and treatment.

Acute Myeloid Leukemia Stem Cells: Origin, Characteristics, and Clinical Implications

Summary: This article reviews the biology of leukemia stem cells (LSCs) in acute myeloid leukemia (AML), focusing on their relationship with the heterogeneous nature of AML and its high relapse rate. The defining functional and phenotypic features of LSCs are discussed, including their surface and metabolic phenotypes that differentiate them from normal hematopoietic stem cells (HSCs). The potential for using this information to improve therapy and address clinical needs is highlighted, as well as the current gaps in the scientific understanding of LSCs.
