Summary: A series of novel atropisomeric diphosphine ligands termed TanPhos with a smaller bite angle were synthesized and designed. TanPhos exhibited high reactivity and enantioselectivity in the rhodium-catalyzed asymmetric hydrogenation of α-dehydro amino ketones, achieving up to 99% yield and 99% ee for a wide range of chiral α-amino ketones.

Summary: This study reports a palladium-catalyzed asymmetric allylic alkylation method that achieves desymmetrization and atroposelectivity. The palladium complex with a chiral phosphoramidite ligand enables the enantioselective transformation of nucleophilic 2-arylresorcinols. Computational studies provide insight into the transition states involving intra- or intermolecular hydrogen bonding, with the energetically favorable transition state matching the observed enantioselectivity. The desymmetrization of symmetric biaryls offers an efficient and underexplored approach to the synthesis of axially chiral molecules.

Remarkably, more than half of this output was achieved in the last twenty years, showcasing Trost's inventiveness and ingenuity. This review provides a brief summary of all total syntheses from the Trost laboratories since 2000, highlighting sixteen particularly notable campaigns in the field.

Summary: Utilizing the pyridinyl moiety as the coordinating group in cobalt-catalysed asymmetric nucleophilic addition/alkylation of secondary phosphine oxides can lead to the efficient synthesis of tertiary phosphine oxides with high yields and high enantiomeric excess, along with broad functional-group tolerance. Mechanistic studies reveal that (R)-secondary phosphine oxides preferentially interact with the cobalt catalysts to produce P-stereogenic compounds.

Summary: The study reported the first successful enantioselective hydrosilylation of cyclic imines promoted by a chiral zinc complex, achieving pharmaceutically relevant enantioenriched 2-aryl-substituted pyrrolidines with excellent enantioselectivities. The synthetic utility of the presented methodology was demonstrated in an efficient synthesis of pharmaceutical drug precursors to Aticaprant and Larotrectinib.

Summary: We synthesized a new asymmetric heptadentate ligand based on the 1,3-diaminopropan-2-ol back-bone. The ligand contains two amide and two pyridine groups attached to the core. Reaction with Zn(ClO4)2.6H2O resulted in the formation of a dinuclear complex. The complex showed pH-dependent phosphatase activity with the highest activity at pH 9.

Summary: A novel dimeric iodo-iridium(iii) complex with an amide moiety within the tetramethylcyclopentadiene ring has been synthesized and characterized. The presence of restricted rotation in the 2-substituted isomer leads to the existence of two distinguishable conformers. The relative acidities of Cp*CONMe2 and Cp* have been compared, and the study also focuses on the decrease in enantiomeric excess during the reduction reaction.

Summary: The isomerism of zinc complex [Zn(L)(2)](2+) with tridentate ligand L was studied using DFT calculations, liquid state NMR, and single-crystal X-ray diffraction. The most stable isomer of [Zn(L)(2)](2+) complex was found to be mer-C-2 symmetric, consistent with the X-ray structure. The cancellation of magnetic equivalence in some nuclei upon coordination to Zn2+ cation was explained by averaging of NMR parameters in the calculation procedure.
