Summary: We report a highly enantioselective desymmetrization reaction for the synthesis of axially chiral biaryl N-oxides by atroposelective C-H iodination using a chiral catalyst coordinated by N-benzoyl-l-phenylalanine. A broad range of products were obtained in high yields (up to 99%) with excellent enantioselectivities (up to 98% ee). The products could be synthesized in gram scale, and one of them was shown to be a powerful organocatalyst in asymmetric allylation reaction. Mechanistic evidence and DFT calculations provide insights into the factors leading to high reactivity and excellent enantiocontrol in this reaction.

Summary: The development of a deconstructive strategy for the first asymmetric synthesis of (-)-thebainone A was described, utilizing an enantioselective C-C bond activation and a C-O bond cleavage reaction. The newly optimized conditions show broad substrate scope and excellent enantioselectivity (up to 99.5:0.5 er). The synthesis of (-)-thebainone A was completed by taking advantage of boron-mediated ether bond cleavage through two complementary routes.

Summary: This study reports the Pd(II)-catalyzed dC(sp(2))-H (ortho) functionalization of phenylalaninol scaffolds directed by picolinamide. Various arylated, alkylated, benzylated derivatives of phenylalaninol scaffolds were successfully synthesized. The hydrolysis of the picolinamide moiety and synthetic utility of the arylated substrates were investigated. The preparation of modified Matijin-Su derivatives using the arylated phenylalaninol compounds was also demonstrated.

Summary: Thiocyanates, a versatile building block in organic synthesis, can be easily accessed through an interrupted Pummerer reaction of sulfoxides. This dealkylative functionalization can proceed under mild conditions by electrophilic activation of the sulfoxide partner, and the resulting thiocyanate can be used as a handle for diversification in a straightforward one-pot procedure.

Summary: A regioselective B(4)-alkenylation method has been developed using o-carboranyl sulfoxonium ylides and alkynes through B(4)-H activation catalyzed by Iridium(I). Sequential B(4)- and B(6)-alkenylation reactions resulted in the synthesis of B(3,5)-dialkenylated o-carboranyl sulfoxonium ylides in one pot. This method allows the introduction of two alkenyl groups at positions 3 and 5 of the carborane, with the sulfoxonium ylide serving as a directing group that can be further functionalized to B-alkenylated o-carboranyl trichloromethyl ketones.

Summary: By using eosin Y as a catalyst, this study demonstrates a photothermally induced decarbonylative aryl C-H arylation with aldehydes. The reaction is simple, solvent-free, and does not require 1,2-dinitrobenzene additive. It also tolerates previously unsuccessful functionalities. In addition to aryl aldehydes, alkyl aldehydes are also compatible. Mechanistic investigations reveal the reaction pathway involving aldehyde decarbonylation, regioselective addition, and oxidation.

Summary: A Rh(III)-catalyzed dehydrogenative annulation and spirocyclization of 2-arylindoles and 2-(1H-pyrazol-1-yl)-1H-indole with maleimides has been developed, providing highly functionalized compounds. The reaction methodology has broad substrate scope, good functional group tolerance, and is simple and scalable. The annulated product of 2-(1H-pyrazol-1-yl)-1H-indole showed significant red-shift and high absorption and emission values compared to 2-phenylindole.

Summary: Regiocontrol is challenging in traditional cycloaddition reactions between unsaturated carbon compounds. This study reports a regio-controllable synthesis of benzocyclobutenes (BCBs) through a palladium-catalyzed double C-H activation of adjacent methylene units in carboxylic acids using bidentate amide-pyridone ligands. This method allows for the synthesis of diversely functionalized BCBs and hetero-BCBs found in drug molecules and bioactive natural products.

Summary: The reactivity of sulfoxonium ylide with allyl carbonates and allyl carbamates diverges. Rh(iii)-catalyzed C-H activation and cyclization of sulfoxonium ylide with ally esters leads to the formation of a cyclopropane-fused tetralone derivative via cascade C-H activation, (4+2) annulation, and cyclopropanation. A similar reaction of sulfoxonium ylide with allyl carbamates forms a C3-substituted indanone derivative through a rare and interesting domino C-H activation and (4+1) annulation in which allyl carbamate acts as a C1-synthon.

Summary: The coronavirus pandemic has resulted in a large number of deaths worldwide, emphasizing the urgent need for effective treatments against COVID-19. This study explores the potential of combining the antitussive drug, noscapine, with antiviral drugs for treating the disease. The researchers found that the noscapine-hydroxychloroquine conjugate showed strong binding to the main protease of SARS-CoV-2, which is crucial for the virus's infection and progression. This combinatorial therapy could be further investigated as a potential treatment for COVID-19.
