An efficient method for the liquid-phase synthesis of 3,5-disubstituted thiohydantoins has been developed. This new class of substituted thiohydantoins was tested as DNA fluorescence probes. Anthracene-substituted thiohydantoin undergoes a change in fluorescence in the presence of DNA. (C) 2011 Elsevier Ltd. All rights reserved.

Cyanine mitochondrial dye with slightly selective cytotoxicity against A549 cancerous cells

Summary: Delocalized lipophilic cations (DLCs) are molecules known to target mitochondria and may have antioxidant or anticancer properties. The novel cytotoxic fluorescent compound 62E2 exhibits cytotoxic and mitotoxic effects at low concentrations, with some selectivity against A549 cancer cells. Its closest analog, F16, has also been described as a potential anticancer agent.

Telomere length regulation by Rif1 protein from Hansenula polymorpha

Summary: This study reports the functional characterization of the Rif1 homologue from the methylotrophic thermotolerant budding yeast Hansenula polymorpha DL-1. The study reveals that Rif1 in H. polymorpha suppresses telomerase-dependent telomere elongation through direct DNA binding and association with the Ku heterodimer. It is also found that Rif1 binds Stn1 and promotes its accumulation at telomeres in H. polymorpha.

A Versatile System for Comparing Methods for Determining Mutations in the TERT Gene Promoter as an Example of the Analysis of Tumor Lesions of the Central Nervous System

Summary: This study compared the methods of real-time PCR and digital droplet PCR for detecting mutations in the promoter region of the TERT gene, showing the advantage of digital droplet PCR in analyzing central nervous system tumor lesions. By analyzing mutations in the TERT gene of human brain tumor cell cultures, the mutant allelic fraction for Grade IV glioblastoma IDH1 was determined for the first time.

Biotinylated Pt(iv) prodrugs with elevated lipophilicity and cytotoxicity

Summary: A design of Pt(iv) prodrugs with tumor cell targeting moieties leading to increased selectivity is of interest. Herein, we designed a novel Pt(iv) prodrugs with COX-inhibitor naproxen, long-chain hydrophobic stearic acid moiety and biotin as axial ligands. We have established that for Pt(iv) prodrugs with biotin and naproxen or stearate in axial position, the lipophilicity rather than biotin receptors expression is the main factor of cytotoxicity. We also monitored the reduction speed of Pt(iv) prodrug 3 with naproxen and biotin in axial positions in A549 cells using XANES and demonstrated that the prodrug gradually releases cisplatin within 20 hours of incubation.

The synthesized compounds were evaluated for their biological activity.

Summary: A series of new organic ligands consisting of two 5-(2-pyridylmethylene)-3,5-dihydro-4H-imidazol-4-one units linked with polymethylene chains of various lengths have been synthesized. These ligands react with copper salts to form mono- and binuclear complexes containing Cu2+ or Cu1+, showing higher cytotoxicity and reasonable selectivity to cancer cell lines.
