Summary: New phthalazone tethered 1,2,3-triazole derivatives 12-21 were synthesized through Cu(I)-catalyzed click reactions. The synthesized compounds were confirmed by spectroscopic tools and elemental analysis. The antiproliferative efficacy of compounds 12-21 against cancer cell lines was evaluated, and compound 16 showed potent activity and selectivity. In silico molecular docking confirmed stable protein-ligand interactions for the effective derivatives.

Summary: A versatile approach was developed for the preparation of porous polystyrene frameworks functionalized with triazolyl moieties. The porous materials were synthesized from PLA-b-P(S-stat-4-AMS) diblock copolymer precursors and characterized by various analytical techniques. Functionalization of the materials with p-tolylacetylene or 2-methyl-3-butyn-2-ol was achieved via copper-catalyzed azide-alkyne cycloaddition, resulting in high yields of the triazole ring.

Summary: Efficient consecutive formation of 1,2,3-triazoles using multiazide platforms has been achieved in this study. A four-step synthesis of tetrakis(triazoles) was successfully performed from a tetraazide platform molecule, based on the unique clickability of the 1-adamantyl azido group. This method was applied to the convergent synthesis of tetrafunctionalized probes in a modular synthetic manner.

Summary: Amino acid-tuned self-assembly is an attractive strategy for constructing functional materials, including nanovaccines for cancer immunotherapy. The top-performing material identified in this study, DA6C1, can significantly enhance cellular uptake of antigens and oligonucleotides, leading to improved activation of dendritic cells. Subcutaneous injection of DA6C1-based nanovaccine can elicit strong immune responses for effective prophylactic and therapeutic effects.

Summary: A novel series of compounds incorporating 1,4-triazole and 1H-tetrazole pharmacophoric motifs linked to the 1,4-benzodiazepine nucleus were synthesized through a domino click reaction. The structures of these compounds were determined using various analytical techniques. A remarkable feature of this approach is the easy access to 1,4-regioisomers of triazole and tetrazole ring incorporated heterocyclic materials through a one-pot strategy using Cu(I) catalyzed cycloaddition. The synthesized compounds showed potential antibacterial activity and were studied using molecular docking techniques.

Summary: A series of tyrosol 1,2,3-triazole derivatives were synthesized and tested for phytotoxic activity on wild poinsettia, fleabane, and tropical spiderwort. While none of the compounds showed activity against two of the weed species, they exhibited strong phytotoxic effects on wild poinsettia, indicating their potential use as tools for managing this competitive weed in agricultural lands.

Summary: Proteolysis Targeting Chimera (PROTAC) is a bifunctional molecule that degrades binding proteins through the ubiquitin-proteasome pathway. Despite its potential in overcoming drug resistance and targeting undruggable targets, PROTAC faces challenges such as poor membrane permeability and bioavailability due to its large molecular weight. In this study, we used intracellular self-assembly to construct tumor-specific PROTACs using small molecular precursors. These precursors, incorporated with azide and alkyne as bioorthogonal groups, reacted with each other under the catalysis of copper ions in tumor tissues, resulting in novel PROTACs. These intracellular self-assembled PROTACs effectively induced degradation of VEGFR-2 and EphB4 in U87 cells, as well as promoting apoptosis and blocking cells in S phase. The high selectivity of these tumor-specific intracellular self-assembled PROTACs was attributed to the high concentration of copper ions in tumor tissue. Additionally, this strategy reduced the molecular weight of PROTACs and improved membrane permeability. These findings greatly expand the application of bioorthogonal reactions in the discovery of novel PROTACs.

Summary: In this study, a low-metal iron-supported catalyst (Fe20/NC-Mg) was reported for the efficient synthesis of quinoxaline compounds, using water as a solvent and without additional bases. The synergistic effect of FeNx site and Mg (OH)2 nanorods in the catalyst played a key role in achieving high yields (82-91%) in 16 examples. The gram-level synthesis and reusability of the catalysts after four cycles demonstrated its industrial application potential.
