A novel method for speciation of Pt in human serum incubated with cisplatin, oxaliplatin and carboplatin by conjoint liquid chromatography on monolithic disks with UV and ICP-MS detection

Conjoint liquid chromatography (CLC) on monolithic convective interaction media (CIM) disks coupled on-line to UV and inductively coupled plasma mass spectrometry (ICP-MS) detectors was used for the first time in speciation analysis of Pt in human serum spiked with Pt-based chemotherapeutics. CIM Protein G and CIM DEAE disks were assembled together in a single housing forming a CLC monolithic column. Such a set-up allows rapid two-dimensional separation by affinity and ion-exchange (IE) modes to be carried out in a single chromatographic run. By applying isocratic elution with Tris-HCl-NaHCO3 buffer (pH 7.4) in the first minute, followed by gradient elution with 1 mol L-1 NH4Cl (pH 7.4) in the next 9 min, immunoglobulins (IgG) were retained by the Protein G disk enabling subsequent separation of unbound Pt from Pt bound to transferrin (Tf) and albumin (HSA) on the CIM DEAE disk. Further elution with acetic acid (AcOH) in the next 3 min allowed separation of Pt associated with IgG. Separated Pt species were quantified by post-column isotope dilution ICP-MS. Pt recovery on the CLC column was close to 100%. In comparison to commonly applied procedures that involve separation of protein peaks by size-exclusion chromatography (SEC) followed by IE separation of metal-based chemotherapeutic fractions bound to serum proteins, the CLC method developed is much faster and simpler. Its sensitivity (LOQs adequate for quantification of all separated Pt species, lower than 2.4 ng Pt mL(-1)), good selectivity and method repeatability (RSD +/- 3%) enabled investigation of the kinetics of interaction of Pt-based chemotherapeutics with serum proteins and the distribution of Pt species in spiked human serum. Pt species present in spiked serum were bound preferentially to HSA. The proportion of Pt associated with IgG and Tf was lower than 13%. Cisplatin and especially oxaliplatin react rapidly with serum proteins, while carboplatin much less. The method developed may be reliably applied in preclinical and clinical studies of the kinetics of the interaction and distribution of different metallodrugs with proteins in blood serum. Differences in Metal Ions Released from Orthodontic Appliances in an In Vitro and In Vivo Setting

Summary: This study assessed the composition of released metal ions from fixed orthodontic appliances in both in vitro and in vivo settings, and found differences in the metal ion content between these two environments. Causation of Oxidative Stress and Defense Response of a Yeast Cell Model after Treatment with Orthodontic Alloys Consisting of Metal Ions

Summary: This study investigated the metal composition and release of metal ions from orthodontic alloys, as well as the oxidative consequences that the metal ions may cause in the cell. The results showed that beta titanium alloy released the fewest metal ions and did not cause oxidative stress or protein damage. Stainless steel and cobalt-chromium alloy, on the other hand, can cause oxidative stress and protein damage at high concentrations. All metal ions from orthodontic alloys alter the activity of antioxidant enzymes in some way.

Numerical mesoscale tissue model of electrochemotherapy in liver based on histological findings

Tumor Radiosensitization by Gene Electrotransfer-Mediated Double Targeting of Tumor Vasculature

Summary: Targeting specific endothelial cell markers involved in different signaling pathways for tumor radiosensitization shows promising potential. We evaluated a plasmid for silencing two prominent targets and explored the therapeutic potential of gene electrotransfer-mediated delivery of this plasmid in an in vivo study. The results demonstrate significant radiosensitization and a good therapeutic potential, making it a promising novel treatment modality for a wide range of tumors.

Comparison of general anesthesia and continuous intravenous sedation for electrochemotherapy of head and neck skin lesions

Electrochemotherapy of Melanoma Cutaneous Metastases in Organ Transplant Recipients: A Systematic Review of Preclinical and Clinical Studies

Summary: Cutaneous melanoma, a highly aggressive form of skin cancer, has seen improved treatment with the development of immune checkpoint inhibitors (ICIs). The tumor microenvironment (TME) plays a crucial role in the success of ICIs, and understanding its interactions is important for optimizing their efficacy. Electrochemotherapy (ECT) has been shown to enhance ICIs by inducing tumor cell death, releasing tumor antigens, and modulating the TME. This review summarizes the knowledge of TME changes after ECT in melanoma treatment and highlights differences in tumor-infiltrating immune cells between immunocompetent and immunosuppressed organisms, as well as the potential benefits of ECT for organ transplant recipients.

HPV-positive murine oral squamous cell carcinoma: development and characterization of a new mouse tumor model for immunological studies

The effectiveness of calcium electroporation combined with gene electrotransfer of a plasmid encoding IL-12 is tumor type-dependent

Summary: This study compared the effectiveness of CaEP and ECT, and investigated the impact of the combination of CaEP and IL-12 GET on tumors. The results showed that CaEP was more effective than ECT in vitro and in the 4T1 tumor model, while the efficacy of CaEP and ECT was similar in the B16-F10 tumor model. Furthermore, the combination of CaEP and IL-12 GET could modify tumor immune cells and vasculature, further enhancing the antitumor effect.

Safety and Feasibility of Vulvar Cancer Treatment with Electrochemotherapy

Summary: Hepatocellular carcinoma (HCC) is a major global health challenge, ranking third in cancer deaths worldwide. This study focuses on targeting molecules with aberrant expression confined to tumor cells, particularly non-coding RNAs (ncRNAs) such as miRNAs, lncRNAs, and circRNAs. These ncRNAs play a significant role in regulating various aspects of HCC, including proliferation, apoptosis, invasion, and metastasis. The review provides the latest state-of-the-art approaches, highlighting key trends and opportunities for advanced and efficacious HCC treatments.

Effects of Electrochemotherapy on Immunologically Important Modifications in Tumor Cells

The results showed that intratumoral administration had better treatment outcomes, with significantly improved local tumor control, longer disease-free interval, and progression-free survival compared to the other routes.

Summary: In this study, a fluorescent biosensing system for high sensitivity detection of MT was established using a new Ag+-stabilized DNA triplex probe (Ag+-SDTP) combined with catalytic hairpin assembly (CHA) and CRISPR/Cas12a signal enhancements. The sensing method is able to selectively detect MT from other non-specific molecules and can achieve low level detection of MT in diluted human serums, demonstrating its potential for monitoring disease-specific MT biomarker at trace levels.

Innovative SALDI mass spectrometry analysis for Alzheimer's disease synthetic peptides detection

CRISPR-empowered electrochemical biosensor for target amplification-free and sensitive detection of miRNA

Summary: We developed a CRISPR-empowered electrochemical biosensor, named PER-E-CRISPR, for amplification-free and sensitive detection of miR-21. The biosensor combines the advantages of CRISPR/Cas13a and primer exchange reaction (PER) for target amplification. Under optimized conditions, the PER-E-CRISPR assay exhibits a linear range from 10-13 to 10-7 M and a limit of detection of 30.2 fM. The biosensor shows excellent performance in actual plasma, indicating promising prospects for miRNA detection in molecular diagnosis.

Application and evaluation of molecular docking for aptamer and small molecular interaction- A case study with tetracycline antibiotics

Summary: Molecular docking (MD) analysis is widely used for studying the interaction between aptamers and small molecules. This study established steady-state aptamers of tertiary structures (SATS) using specific aptamers of tetracycline antibiotics as docking models. Molecular docking results revealed multiple binding sites in the SATS of aptamers, with significant variations in binding free energy (BFE) and docking score (DS). The study also proposed a method for MD analysis based on SATS, providing insights into binding mode and predicting binding sites.

The construction of highly selective surface molecularly imprinted polymers based on Cu(II) coordination for the detection of bisphenol A

Summary: In this study, a highly selective molecularly imprinted polymer Cu-MIPs@CS was designed and synthesized for bisphenol A (BPA). Cu-MIPs@CS showed significantly enhanced imprinting factor and selective factor towards BPA compared to MIPs@CS. It also exhibited superior discrimination ability between BPA and its structural analogue. Additionally, Cu-MIPs@CS were successfully applied as a solid phase extraction adsorbent for the determination of BPA in drinking water samples.

A simple joint detection platform for high-throughput single-cell heterogeneity screening
