Rapid assessment of endpoint antioxidant capacity of red wines through microchemical methods using a kinetic matching approach

Antioxidant capacity of food samples is usually assessed by different analytical methods, however the results attained even for the same method are strongly dependent on the selected reaction time and also on the standard compound used. To tackle this problem, we propose here a kinetic matching approach, associated to the conversion of results into equivalents of a common standard compound, as a universal way for expression of results. The methodology proposed was applied to methods based on different chemistries (Folin-Ciocalteu (F-C), CUPRAC, DPPH center dot and ABTS(center dot+) assays) and red wines (n=40) were chosen as a model of complex food sample. For implementation of the kinetic matching approach, the standard phenolic mixture (caffeic acid, (+)-catechin, hesperetin, morin and (-)-epigallocatechin gallate) was chosen for calibration in F-C, CUPRAC and DPPH center dot assays, while tannic acid was suitable for ABTS(center dot+) assay. Results showed that, for all methods, there was no statistical difference between results attained by the kinetic matching approach (after < 10 min of reaction) and that at endpoint conditions (after 60 to 300 min). The repeatability and the reproducibility of the kinetic matching approach was < 4.5%, for all antioxidant assays. The sample throughput increases from < 18 (endpoint measurements) to > 108 h(-1) using the proposed kinetic approach. Moreover, we have established here a way of converting results to equivalents of a common standard, providing values independent of its kinetic profile, by using the ratio between calibration sensitivities performed at endpoint conditions. A data mining tool for untargeted biomarkers analysis: Grapes ripening application

Sample preparation and chromatographic methods for the determination of protein-bound uremic retention solutes in human biological samples: An overview

Summary: This review addresses the monitoring and evaluation of protein-bound uremic retention solutes in biological fluids, such as indole-3-acetic acid, indoxyl sulfate, p-cresol, and p-cresol sulfate, which have been associated with the development of kidney, cardiovascular, and bone toxicities. The review discusses conventional and advanced sample treatment procedures and chromatographic analytical methods for quantification of these compounds, with a particular focus on plasma, serum, and urine samples. There is a trend towards using liquid chromatography-tandem mass spectrometry for analysis, requiring protein precipitation, solid-phase extraction, and/or dilution prior to sample analysis. Miniaturized methods using microplate devices are recommended for sustainability.

Chemometric-assisted surface-enhanced Raman spectroscopy for metformin determination using gold nanoparticles as substrate

Summary: A fast, simple, and reliable method for determination of metformin was developed by coupling surface-enhanced Raman spectroscopy (SERS) with chemometric methods. The method utilized a portable Raman spectrometer and citrate stabilized gold nanoparticles (AuNPs) as substrate, resulting in improved sensitivity. Principal component analysis (PCA) and partial least squares (PLS) regression were used for data analysis. Optimization of the PLS model led to RMSECV and R2CV values of 0.42 mg/L and 0.94, respectively. Validation of the optimized PLS regression model with commercial pharmaceutical samples yielded good results in terms of R2P (0.97), RE (4.54%), and analytical sensitivity (2.13 mg/L).

Rational design of magnetoliposomes for enhanced interaction with bacterial membrane models

Summary: This study aims to develop and optimize the production of PEGylated magnetoliposomes (MLPs@PEG) by encapsulating superparamagnetic iron oxide nanoparticles (SPIONs) within fusogenic liposomes. The optimized MLPs@PEG has a mean size of 182 nm, polydispersity index (PDI) of 0.19, and SPIONs encapsulation efficiency (%EE) around 76%. Cytocompatibility assays demonstrate that the MLPs@PEG exhibits no toxicity in fibroblasts and no hemolytic effect at safe lipid and iron concentrations.

Exploiting Kinetic Features of ORAC Assay for Evaluation of Radical Scavenging Capacity

Summary: The analysis and interpretation of data from ORAC assays are difficult. This study compares the expression of ORAC values as AUC and lag time. It shows that some antioxidant compounds have radical scavenging reactions that extend beyond the lag time. Ethanol delays the reaction of FL and antioxidant compounds with free radicals. The choice of calibrator compounds is important to prevent biased estimation of antioxidant capacity.

Innovative SALDI mass spectrometry analysis for Alzheimer's disease synthetic peptides detection

CRISPR-empowered electrochemical biosensor for target amplification-free and sensitive detection of miRNA

The biosensor combines the advantages of CRISPR/Cas13a and primer exchange reaction (PER) for target amplification. Under optimized conditions, the PER-E-CRISPR assay exhibits a linear range from 10-13 to 10-7 M and a limit of detection of 30.2 fM. The biosensor shows excellent performance in actual plasma, indicating promising prospects for miRNA detection in molecular diagnosis.

Application and evaluation of molecular docking for aptamer and small molecular interaction- A case study with tetracycline antibiotics

Summary: Molecular docking (MD) analysis is widely used for studying the interaction between aptamers and small molecules. This study established steady-state aptamers of tertiary structures (SATS) using specific aptamers of tetracycline antibiotics as docking models. Molecular docking results revealed multiple binding sites in the SATS of aptamers, with significant variations in binding free energy (BFE) and docking score (DS). The study also proposed a method for MD analysis based on SATS, providing insights into binding mode and predicting binding sites.

The construction of highly selective surface molecularly imprinted polymers based on Cu(II) coordination for the detection of bisphenol A

Summary: In this study, a highly selective molecularly imprinted polymer Cu-MIPs@CS was designed and synthesized for bisphenol A (BPA). Cu-MIPs@CS showed significantly enhanced imprinting factor and selective factor towards BPA compared to MIPs@CS. It also exhibited superior discrimination ability between BPA and its structural analogue. Additionally, Cu-MIPs@CS were successfully applied as a solid phase extraction adsorbent for the determination of BPA in drinking water samples.

A simple joint detection platform for high-throughput single-cell heterogeneity screening
