The Evolution of Defense Mechanisms Correlate with the Explosive Diversification of Autodigesting Coprinellus Mushrooms (Agaricales, Fungi)

Bursts of diversification are known to have contributed significantly to the extant morphological and species diversity, but evidence for many of the theoretical predictions about adaptive radiations have remained contentious. Despite their tremendous diversity, patterns of evolutionary diversification and the contribution of explosive episodes in fungi are largely unknown. Here, using the genus Coprinellus (Psathyrellaceae, Agaricales) as a model, we report the first explosive fungal radiation and infer that the onset of the radiation correlates with a change from a multilayered to a much simpler defense structure on the fruiting bodies. We hypothesize that this change constitutes a key innovation, probably relaxing constraints on diversification imposed by nutritional investment into the development of protective tissues of fruiting bodies. Fossil calibration suggests that Coprinellus mushrooms radiated during the Miocene coinciding with global radiation of large grazing mammals following expansion of dry open grasslands. In addition to diversification rate-based methods, we test the hard polytomy hypothesis, by analyzing the resolvability of internal nodes of the backbone of the putative radiation using Reversible-Jump MCMC. We discuss potential applications and pitfalls of this approach as well as how biologically meaningful polytomies can be distinguished from alignment shortcomings. Our data provide insights into the nature of adaptive radiations in general by revealing a deceleration of morphological diversification through time. The dynamics of morphological diversification was approximated by obtaining the temporal distribution of state changes in discrete traits along the trees and comparing it with the tempo of lineage accumulation. We found that the number of state changes correlate with the number of lineages, even in parts of the tree with short internal branches, and peaks around the onset of the explosive radiation followed by a slowdown, most likely because of the decrease in available niches.

Summary: Plant pathogenic fungi cause significant losses in crop production worldwide. Developing new strategies for plant and crop protection is crucial due to the resistance of these fungi to approved chemical fungicides. A study investigated the potential of using extracellular antifungal proteins (AFPs) and their peptide derivatives (PDs) as combinatorial biofungicides for plant and crop protection. The study found that the combination of AFPs and PDs can effectively inhibit the growth of various fungi and mitigate the symptoms of fungal infection on tomato plants.

Selection and Characterization of a Bacillus Strain for Potential Application in Industrial Production of White Button Mushroom (Agaricus bisporus)

Summary: This study isolated and examined bacterial strains from mushroom casing material for their potential use as biocontrol agents against mold pathogens of white button mushroom. The selected Bacillus velezensis strain showed effective control against the pathogen, and even increased the crop yield of A. bisporus. These findings provide a potential method for the biological and integrated treatment in Agaricus cultivation.

Differential Gene Expression of Mucor lusitanicus under Aerobic and Anaerobic Conditions

Summary: This study reveals significant changes in gene expression of Mucor lusitanicus under aerobic and anaerobic conditions through transcriptomic analysis. Carbohydrate metabolism and transporter genes, especially sugar transporters, are found to be affected. Moreover, transcripts related to amino acid transport and metabolism, lipid transport and metabolism, and energy production and conversion are downregulated under anaerobic conditions.

Quinone binding site in a type VI sulfide:quinone oxidoreductase

Summary: This study reports the characteristics of the quinone binding site (QBS) of a type VI sulfide:quinone oxidoreductases (SQR) from the photosynthetic purple sulfur bacterium Thiocapsa roseopersicina. The study identifies the amino acids near the enzyme-bound quinone ring and their interaction with the quinone head group. Mutant experiments further confirm the importance of these interactions.

Pretreatment of lignocellulosic biogas substrates by filamentous fungi

Summary: This study investigated the decomposition of lignocellulosic plant biomass by four filamentous fungi. It was found that the Penicillium aurantiogriseum strain exhibited excellent hydrolytic enzyme production and generated the highest biogas yield from partially degraded substrates. Corn stover was the most suitable substrate for biomass decomposition and biogas production.

This paper presents the physical-chemical properties of the homologues in mice and humans, providing insights into the CYB561 protein family.

Hard nut to crack: Solving the disulfide linkage pattern of the Neosartorya (Aspergillus) fischeri antifungal protein 2

Summary: Due to the fast spread of drug resistance, only a limited number of drugs can be used to treat fungal infections. Therefore, there is an urgent need to develop new antifungal treatment strategies. The NFAP2 protein secreted by the mold Neosartorya shows promise as a template for future antifungal drug design, but it is difficult to determine its disulfide linkage pattern using traditional mass spectrometry-based methods.

Scale-up of Aflatoxin Purification by Centrifugal Partition Chromatography

Summary: Aflatoxins (AFs) are secondary metabolites that cause diseases in animals and humans. Regulations set concentration limits for AFs in EU food and feed products, requiring pure forms for reference standards. Improvements were made to a liquid-liquid chromatographic method to enhance purification and increase the yield of pure AFs. Through a scale-up process, approximately 2.2 g of AFs could be purified using 8.2 L of solvent in a 250 mL rotor, while 7.8 g of AFs could be prepared using 31 L of solvent in a 1000 mL rotor.

Advantages of Multiplexing Ability of the Orbitrap Mass Analyzer in the Multi-Mycotoxin Analysis

Reversal of Multidrug Resistance by Symmetrical Selenoesters in Colon Adenocarcinoma Cells

Summary: Recently, the anticancer activity of symmetrical selenoesters was investigated in sensitive and resistant human colon adenocarcinoma cells. The compounds showed synergistic interaction with doxorubicin and exhibited ABCB1 inhibitory and apoptosis-inducing effects. Transcriptome analysis revealed that symmetrical selenoesters may influence the redox state of cells and interfere with metastasis formation.

Development of a Multicomponent Microbiological Soil Inoculant and Its Performance in Sweet Potato Cultivation

Summary: The cultivation and consumption of sweet potato are increasing globally, but the use of chemical fertilizers and pesticides may cause pollution. Therefore, there is a need for environmentally-friendly biological solutions. This study developed a microbial inoculant from multiple microorganisms and tested its application in sweet potato cultivation, resulting in increased yield. This is the first successful application of a fungal-bacterial consortium in sweet potato cultivation.

Regulation of the methanogenesis pathways by hydrogen at transcriptomic level in time

Summary: This study investigated the biomethane formation from 4 H-2 + CO2 by two methanogens, Methanocaldococcus fervens and Methanobacterium thermophilum. The research aimed to understand the regulation of biomethane biosynthesis by H-2. It was found that the autotrophic methanogen M. fervens could be easily controlled by H-2/CO2, while the heterotrophic methanogen M. thermophilum was less sensitive to H-2/CO2, suggesting the importance of the biomethane-generating methanogenic community in renewable excess electricity conversion and storage systems.

Structure-activity correlations for peptaibols obtained from clade Longibrachiatum of Trichoderma: A combined experimental and computational approach

Summary: This study investigated the production of peptaibols by various Trichoderma strains and their correlation with structure-activity relationships. The results showed that T. longibrachiatum f. bissettii SZMC 12546 exerted the lowest minimum inhibitory concentrations (MIC) against Gram-positive bacteria and also inhibited the plant pathogenic Gram-negative Rhizobium radiobacter. Molecular dynamics simulations revealed that the folding dynamics of Group A peptaibols were more restricted and had well-folded helical conformations, which may contribute to their bioactivity against bacteria.
