In the last few years matrix assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) has been increasingly studied and applied for the identification and typing of microorganisms. Very recently, MALDI-TOF MS has been introduced in clinical routine microbiological diagnostics with marked success, which is remarkable considering that not long ago the technology was generally seen as being far from practical application. The identification of microbial isolates by whole-cell mass spectrometry (WC-MS) is being recognized as one of the latest tools forging a revolution in microbial diagnostics, with the potential of bringing to an end many of the time-consuming and man-power-intensive identification procedures that have been used for decades. Apart from applications of WC-MS in clinical diagnostics, other fields of microbiology also have adopted the technology with success. In this article, an over-view of the principles of MALDI-TOF MS and WC-MS is presented, highlighting the characteristics of the technology that allow its utilization for systematic microbiology. Cyanobacteria and Cyanotoxins in a Changing Environment: Concepts, Controversies, Challenges

Ingrid Chorus, Jutta Fastner, Martin Welker

Summary: Climate change may promote cyanobacterial blooms in certain waterbodies; controlling blooms and toxin concentrations require reducing phosphorus and nitrogen concentrations; health risks depend on whether concentrations exceed WHO cyanotoxin guideline values.

Comment on Models predict planned phosphorus load reduction will make Lake Erie more toxic

Jef Huisman, Elke Dittmann, Jutta Fastner, J. Merijn Schuurmans, J. Thad Scott, Dedmer B. Van de Waal, Petra M. Visser, Martin Welker, Ingrid Chorus

Summary: Hellweger et al. suggest that phosphorus limitation will lead to higher concentrations of cyanobacterial toxins in lakes. However, their assumptions about molecular, physiological, and ecological mechanisms lack support and contradict other studies. It is concluded that their take-home message about increased toxicity in Lake Erie due to phosphorus load reduction is fundamentally flawed.

Cyanotoxins associated with macrophytes in Berlin (Germany) water bodies-Occurrence and risk assessment

Jutta Fastner, Jonna Teikari, Anja Hoffmann, Antje Kohler, Sebastian Hoppe, Elke Dittmann, Martin Welker

Summary: Fatal dog poisoning in Tegeler See, Berlin, Germany, caused by neurotoxic cyanobacteria associated with aquatic macrophytes has raised concerns about human exposure, especially for children, to cyanotoxins during recreational activities. A study conducted from 2017 to 2021 analyzed 398 macrophyte samples and 463 water samples from bathing sites in 19 Berlin lakes, detecting cyanotoxins in 66% of macrophyte samples and 50% of water samples. Anatoxins were the most frequently detected toxin group in macrophyte samples (58%), while cylindrospermopsins were most prevalent in water samples (41%). The study also identified the presence of anatoxin-producing cyanobacteria in the water moss Fontinalis antipyretica. Based on toxicological evaluations, a critical anatoxin content of 3 μg/g fresh weight in macrophyte samples is proposed.
