Self-Restrained B Cells Arise following Membrane IgE Expression

Among immunoglobulins (Igs), IgE can powerfully contribute to antimicrobial immunity and severe allergy despite its low abundance. IgE protein and gene structure resemble other Ig classes, making it unclear what constrains its production to thousandfold lower levels. Whether class-switched B cell receptors (BCRs) differentially control B cell fate is debated, and study of the membrane (m) IgE class is hampered by its elusive in vivo expression. Here, we demonstrate a self-controlled mIgE(+) B cell stage. Primary or transfected mIgE(+) cells relocate the BCRs into spontaneously internalized lipid rafts, lose mobility to chemokines, and change morphology. We suggest that combined proapoptotic mechanisms possibly involving Hax1 prevent mIgE(+) memory lymphocyte accumulation. By uncoupling in vivo IgE switching from cytokine and antigen stimuli, we show that these features are independent from B cell stimulation and instead result from mIgE expression per se. Consequently, few cells survive IgE class switching, which might ensureminimal long-term IgE memory upon differentiation into plasma cells.

Neutrophil function and bactericidal activity against Staphylococcus aureus after cardiac surgery with cardiopulmonary bypass

Summary: This study aimed to investigate the impact of cardiopulmonary bypass (CPB) on neutrophil functions. The results showed that CPB induced a systemic inflammation with an increase in circulating mature neutrophils after surgery. Neutrophil lifespan was significantly increased and functions such as NET release and ROS production were enhanced after CPB, while bacteria killing and phagocytosis were not impacted. These data suggest that CPB recruits mature neutrophils via a demargination process and neutrophils remain efficient post-CPB without contributing to immunosuppression.

Summary: IgA autoantibodies may play a role in autoimmune or chronic inflammatory diseases. A novel IgA autoimmune blistering model using genetically modified mice showed that IgA anti-collagen XVII could activate neutrophils and cause tissue damage, while treatment with anti-Fc alpha RI monoclonal antibodies prevented and resolved the disease.

Roles of G4-DNA and G4-RNA in Class Switch Recombination and Additional Regulations in B-Lymphocytes

Summary: Mature B cells utilize class switch recombination (CSR) to diversify immunoglobulin (Ig) production, facilitated by activation-induced deaminase (AID) targeting cytosines in transcribed targeted S regions. G-rich sequences in S regions form canonical G-quadruplexes (G4s) DNA structures, favoring CSR. Small molecules interacting with G4-DNA (G4 ligands) can regulate CSR either positively or negatively. The specific regulatory role of G4 structures in CSR and lymphomagenesis remains a major issue in understanding immune responses.

Summary: This study characterizes the phenotype, transcriptome, life cycle, and function of human tonsil macrophages, and identifies three subtypes with distinct features. CD36(hi) macrophages are related to monocytes, CD36(lo) macrophages have features of embryonic origin, and CD36(int) macrophages display mixed characteristics. Functionally, CD36(hi) macrophages are specialized in stimulating T follicular helper cells by producing Activin A. Stromal cell-derived TNF-alpha is identified as the inducer of Activin A secretion.

Demultiplexing Ig repertoires by parallel mRNA/DNA sequencing shows major differential alterations in severe COVID-19

Summary: To investigate the immune response architecture in COVID-19 patients and understand the factors that can cause or prevent ARDS, we analyzed the B cell responses using flow cytometry and Ig repertoire analysis. Flow cytometry with FlowSOM analysis revealed changes associated with COVID-19 inflammation, including an increase in double-negative B cells and ongoing plasma cell differentiation. We also found the expansion of two disconnected B cell repertoires. Analysis of Ig repertoire patterns showed an early expansion of IgG1 clonotypes with atypical CDR3 characteristics, which correlated with ARDS severity. Additionally, we observed a convergent response with progressively increasing somatic hypermutation and normal or short CDR3 in anti-SARS-CoV-2 clonotypes, which persisted until the memory B cell stage after recovery.

Unique repetitive nucleic acid structures mirror switch regions in the human IgH locus

Summary: Ig genes have the unique ability to be reshaped in B lymphocytes after encountering specific antigens, resulting in improved affinity and antibody production. Ig class switching, an important change in this process, is mediated by DNA repeats and DNA lesions. The discovery of IgH-like DNA repeats in the human IgH locus suggests their regulatory role in antibody production.
