Summary: This study found that EPC transplantation increased the number of M2 microglia/macrophage-phagocytizing apoptotic cells and the expression of CR3. In cultured microglia, EPC-conditioned medium or cultured EPCs increased microglial migration and phagocytosis, and upregulated CR3 expression. Additionally, the CR3 agonist leukadherin-1 reduced brain atrophy and neurological deficits and increased synaptophysin and PSD-95 expression.

Summary: Phosphodiesterase 4 inhibitors (PDE4-I), such as roflumilast, have been shown to have neuroprotective effects by increasing cAMP levels. In this study, it was investigated whether roflumilast can protect the blood-brain barrier (BBB) in the hippocampus after transient global cerebral ischemia (TGCI) in rats. The results showed that roflumilast attenuated BBB disruption and restored endothelial nitric oxide synthase (eNOS) levels in the CA1 hippocampal area. It also increased the levels of B2 cell lymphoma (BcL-2) and neuron-glial antigen-2 (NG2) after global ischemia in rats.

Summary: The vascular endothelium plays crucial roles in maintaining cardiovascular homeostasis, regulating vascular tone, and processing biochemical substances. Dysfunction of endothelial cells may lead to chronic age-related cardiovascular diseases such as ischemic heart disease. Studies have shown that flavonoid compound quercetin, found in various food sources, may help mitigate the risk factors of endothelial dysfunction such as atherosclerosis and hypertension.

Summary: Pulmonary arterial hypertension (PAH) is a severe disease with high mortality even under treatment. Research has shown that the nitric oxide pathway plays a key role in PAH pathophysiology, with many drugs targeting this pathway. Novel compounds targeting on NO pathway hold great potential for future therapy, and understanding interactions with other pathways could lead to new drug discoveries for better clinical outcomes.

Summary: PDE4 inhibitor Roflumilast protects against neuronal damage caused by immune ischemia and activates Notch1/Hes1 signaling, leading to increased expression of TJ and AJ proteins and maintenance of blood-brain barrier integrity. Roflu also improves neurobehavioral outcomes and ameliorates BBB disruption in rats.

Summary: In stable patients with pulmonary arterial hypertension, pulmonary rehabilitation is an effective non-pharmacological treatment. This study aimed to explore the effects of endurance exercise training on vascular function in these patients. The results showed that training promoted mechanisms of damage/repair in favor of repair, leading to potential improvements in hemodynamics and clinical response.

Summary: This study evaluated the efficacy and safety of switching from phosphodiesterase-5 inhibitors (PDE-5is) to riociguat in patients with pulmonary arterial hypertension (PAH). The results showed that riociguat was more effective than PDE-5is in improving hemodynamic parameters, 6-minute walking distance (6MWD), World Health Organization functional class (WHO-FC), and cardiac index (CI). No adverse events were observed during the switch.

Summary: Phosphodiesterase 7 (PDE7) is an enzyme responsible for degrading cAMP, an important cellular messenger, making the search for potential PDE7 inhibitors a hot topic in treating neurodegenerative and inflammatory diseases. S14, a potential PDE7 inhibitor, has shown promising results in studies, while polymeric nanoparticles loaded with S14 have been developed for targeted drug delivery and controlled release. These advancements aim to improve the pharmacokinetic properties of this interesting PDE7 inhibitor.

Summary: An umbrella review was conducted to investigate the function of sildenafil on diseases other than urogenital system. The study found that sildenafil played an important role in many fields, particularly in vascular protection. This finding provides strong evidence for further expansion of sildenafil utilization in other diseases.

Summary: Hypertensive disorders of pregnancy (HDP) are associated with increased risk of cardiovascular disease (CVD). This study found a 2.4-fold increased risk of hypertension 10 years after HDP. Differences in noninvasive measures of cardiovascular risk were primarily driven by the diagnosis of hypertension, suggesting that the long-term risk of CVD after HDP may be mainly attributed to the development of hypertension.

Summary: Dkk3 plays a crucial role in cardiovascular homeostasis, and its deficiency leads to hypertension and impaired vascular function, which can be rescued by restoring Dkk3 expression. This effect is achieved through promoting VEGF expression and activating the VEGF/Akt/eNOS hypotensive pathway.

Summary: CRTC1 in SF1 cells is a sensitive factor for high-fat diet-induced obesity, necessary for normal appetite regulation. It is primarily expressed in the brain, playing an important role in the ventromedial hypothalamus. RNA sequencing analysis of microdissected ventromedial hypothalamus samples revealed that the loss of CRTC1 significantly altered the expression levels of certain genes.
