Summary: Translating cell therapies into clinical practice requires robust production protocols to optimize and standardize cell manufacture and cryopreservation in compliance with regulations. Two phase I clinical trials were conducted on ALS and SPMS patients using human neural stem cells, produced from brain tissue samples extracted from fetuses. Rigorous quality control measures were implemented to ensure safety, stability, and reproducibility of the cells used in the trials.

Summary: The European Bank for induced pluripotent Stem Cells (EBiSC) is a non-profit project that aims to provide storage, quality control, and distribution services for human iPSCs for research worldwide. Through collaboration with the human pluripotent stem cell registry (hPSCreg®), EBiSC has successfully standardized the management of cell lines and associated data, providing a model for other iPSC repositories to enhance the FAIRness of iPSC research globally.

Summary: MAT(2) aligns cells in the manifold space using a deep neural network with a contrastive learning strategy, defining cell triplets based on known cell type annotations to produce a more robust consensus manifold and reconstructing batch-effect-free gene expression, which helps to annotate cell types more effectively.

Summary: This study examined the impact of differential centrifugation and freeze/thaw on the profiles of extracellular vesicles (EVs) and cell-free messenger RNA (cf-mRNA) in plasma using flow cytometric analysis. The results revealed that the differential centrifugation and post-freeze/thaw processing affected the EV subpopulations and cf-mRNA levels. Freezing plasma containing residual platelets resulted in irreversible ex vivo generation of EV subpopulations and cf-mRNA transcripts.

Summary: The MEASURE study aims to standardize the tumorigenicity evaluation of hPSC-derived CTPs, finding that the HEC assay and MACS can improve sensitivity to residual undifferentiated hPSCs. Multisite experiments confirmed the effectiveness of these methods in detecting low levels of undifferentiated cells.

Summary: The study highlights the important role of MSC-derived extracellular vesicles (MSC-EVs) in hematopoietic regulation and immune response after transplantation. MSC-EVs directly fuse with hematopoietic stem cell membranes, affecting their fate, and regulate the proliferation and differentiation of hematopoietic stem cells through the release of various bioactive factors.

Summary: Cell therapies are a promising treatment option for various diseases, and standards for most aspects of the cell therapy lifecycle have been established, but there are limited standardized protocols regarding the day-of infusion. Variations were identified in facility layout, product thawing location, and cell therapy lab location, which influenced the time from thaw to infusion. Opportunities for standardizing and streamlining the infusion workflow were identified to facilitate adoption of cell therapies at more hospitals.

Summary: Stem cells have shown great potential for tissue regeneration and repair due to their unlimited self-renewal and differentiation capabilities. However, the complex interactions between niches and stem cells, along with the limitations of current systems or methods, pose challenges to the multi-faceted applications of stem cells. Emerging evidence suggests that synchrotron infrared microspectroscopy may offer a promising non-invasive technology for stem cell research.

Summary: Stem cell pools are homogeneous cell populations that possess self-renewal and differentiation potential. While they exhibit limited heterogeneity during homeostasis, their plasticity becomes apparent under stress, leading to changes in phenotype, constitution, metabolism, and function. Manipulating these factors offers better control of stem cell behavior and has the potential to revolutionize regenerative medicine.
