Identification of Heme Oxygenase 1 (HO-1) as a Novel Negative Regulator of Mobilization of Hematopoietic Stem/Progenitor Cells

Activation of complement cascade (ComC) play and important role in mobilization of hematopoietic stem/progenitor cells (HSPCs) from bone marrow (BM) into peripheral blood (PB). While there are vast experimental data on the mechanisms and factors that induce or promote mobilization of HSPCs, there is relatively less data on negative regulators of this process. We demonstrate for the first time that heme oxygenase-1 (HO-1) that has a well-documented anti-inflammatory potential plays an important and heretofore unrecognized role in retention of HSPCs in BM niches by i) modulating negatively activation of mobilization promoting ComC, ii) maintaining stromal derived factor-1 (SDF-1) level in the BM microenvironment and iii) attenuating chemotactic responsiveness of HSPCs to SDF-1 and sphingosine-1 phosphate (S1P) gradients in PB. Furthermore, our data showing a positive mobilizing effect by a non-toxic small-molecule inhibitor of HO-1 (SnPP) suggest that blockade of HO-1 would be a promising strategy to facilitate mobilization of HSPCs. Further studies are also needed to evaluate better the molecular mechanisms responsible for the potential effect of HO-1 in homing of HSPCs after transplantation.

Summary: Very small embryonic like stem cells (VSELs) are dormant stem cells deposited during embryogenesis in various tissues, including bone marrow. They are released and circulate in peripheral blood at a low level, but increase in number in response to stressors and tissue/organ damage. This study evaluates CD34(+)Lin(-)CD45(-) and CD133(+)Lin(-)CD45(-) VSELs derived from umbilical cord blood, comparing their molecular and proteomic characteristics.

Summary: Hematopoietic stem/progenitor cells (HSPCs) express receptors for complement cascade (ComC) cleavage fragments C3a and C5a and respond to inflammation-related cues by sensing PAMPs and DAMPs. The activation of Nox2-ROS-Nlrp3 inflammasome by ComC-derived C3a and C5a plays a role in regulating the responsiveness of HSPCs to stress. Additionally, ComC expressed in HSPCs, known as the complosome, may also contribute to regulating hematopoiesis.

This approach shows promise for future human studies during the acute phase of ischemic cardiac injury.

Summary: We discovered that sterile inflammation in the bone marrow plays a crucial role in the mobilization, homing, and engraftment of hematopoietic stem/progenitor cells (HSCPs). Extracellular adenosine (eAdo), generated from the conversion of extracellular adenosine triphosphate (eATP), affects HSPCs trafficking by activating the A(2B) receptor. By upregulating heme oxygenase-1 (HO-1), an inhibitor of Nlrp3 inflammasome, the A(2B) receptor inhibition promotes cell migration and proliferation. Blocking the A(2B) receptor could potentially enhance HSPCs mobilization and hematopoietic reconstitution.
