Summary: Changes in cellular identity can drive malignant progression and resistance to therapy in lung adenocarcinoma. Loss of FoxA1/2 impairs tumorigenesis in NKX2-1-positive LUAD and leads to collapse of a dual-identity state, driving alternative cellular identity programs associated with non-proliferative states.

Summary: This study reveals the regulatory role of m6A modification on the breast cancer-associated lncRNA HOTAIR. Methylated A783 on HOTAIR interacts with the m6A reader YTHDC1, promoting chromatin association of HOTAIR and driving proliferation and invasion of TNBC cells. The findings demonstrate how modification of a single base in an lncRNA can lead to distinct gene regulation mechanisms and cancer-associated phenotypes.

Summary: In this study, a prognostic risk model for lung adenocarcinoma (LUAD) was constructed using long non-coding RNAs (lncRNAs) related to m5C RNA methylation. A total of 16 lncRNAs significantly associated with overall survival were identified. The study concluded that the m5C-lncRNA signature score was closely related to overall survival based on the constructed risk model and experimental validation.

Summary: ATP-dependent chromatin remodeling complexes, particularly the SWI/SNF complex, play crucial roles in regulating gene expression and have been found to be mutated in a significant percentage of human cancers, including pancreatic ductal adenocarcinoma (PDA). Research has shown that the subunits of the SWI/SNF complex in PDA have context-dependent roles in tumorigenesis, with potential therapeutic implications for targeting their oncogenic or tumor-suppressive properties in PDA. Further understanding of the precise roles of SWI/SNF complex subunits in PDA is necessary for the development of novel therapeutic strategies against this lethal type of cancer.

Summary: HOTAIR, an oncogenic lncRNA, regulates cell processes such as growth and apoptosis by recruiting PRC2 and LSD1 complexes. A quantitative proteomics analysis revealed differential expression of 224 nucleoproteins after HOTAIR inhibition, suggesting potential targets. Further studies on PCNA showed regulation of its expression by HOTAIR, providing novel insights into its mechanism in cancer cells.

Summary: MACC1 expression is associated with poor prognosis in patients with colon adenocarcinoma (COAD), and its upstream miRNA is miR-642a-5p, while the long non-coding RNA (lncRNA) ZFAS1 may serve as its upstream regulator. Additionally, MACC1 expression is positively correlated with tumor immune cell infiltration and biomarker levels of immune cells.

Summary: Cystine uptake promotes GPX4 synthesis by activating mTORC1, sensitizing cancer cells to ferroptosis. Inhibiting mTORC1 decreases GPX4 protein levels and enhances the effectiveness of ferroptosis inducers in cancer treatment.

Summary: The study reveals the mechanism of ferroptosis induced by inhibiting GPX4 in cancer cells, as well as the potential pathways to enhance or inhibit ferroptosis by intervening in relevant metabolic pathways. Therapeutic strategies targeting different levels of GPX4 expression could be a promising new approach for cancer treatment.

Summary: The study reveals the critical role of ferroptosis in p53-mediated tumor radiosensitivity, promoting lipid peroxidation and cell death by inhibiting SLC7A11. P53 deficiency leads to radioresistance in cancer cells or tumors, while ferroptosis inducers that inhibit SLC7A11 significantly enhance tumor sensitivity to radiotherapy.
