Delivery of a mucin domain enriched in cysteine residues strengthens the intestinal mucous barrier

A weakening of the gut mucous barrier permits an increase in the access of intestinal luminal contents to the epithelial cells, which will trigger the inflammatory response. In inflammatory bowel diseases, there is an inappropriate and ongoing activation of the immune system, possibly because the intestinal mucus is less protective against the endogenous microflora. General strategies aimed at improving the protection of the intestinal epithelium are still missing. We generated a transgenic mouse that secreted a molecule consisting of 12 consecutive copies of a mucin domain into its intestinal mucus, which is believed to modify the mucus layer by establishing reversible interactions. We showed that the mucus gel was more robust and that mucin O-glycosylation was altered. Notably, the gut epithelium of transgenic mice housed a greater abundance of beneficial Lactobacillus spp. These modifications were associated with a reduced susceptibility of transgenic mice to chemically induced colitis. Furthermore, transgenic mice cleared faster Citrobacter rodentium bacteria which were orally given and mice were more protected against bacterial translocation induced by gavage with adherent-invasive Escherichia coli. Our data show that delivering the mucin CYS domain into the gut lumen strengthens the intestinal mucus blanket which is impaired in inflammatory bowel diseases.

Summary: A preliminary ethnopharmacological survey was conducted in French Polynesia, focusing on the most cited plants used for treating infectious diseases. The study identified Syzygium malaccense as a potential plant species for further analysis, which led to the discovery of seven alkyl-salicylic acids described for the first time in this species. These compounds showed interesting antimicrobial activity against Gram+ bacteria and no toxicity on hepG2 cells. The findings suggest the therapeutic potential of S. malaccense against infectious diseases.

Outcome of long gap esophageal atresia at 6 years: A prospective case control cohort study

Summary: This study evaluated the outcomes of long gap esophageal atresia (EA) compared to non-long gap EA/tracheo-esophageal fistula (TEF) at the ages of 1 and 6 years. The study also assessed whether the initial treatment (delayed primary anastomosis of native esophagus vs. esophageal replacement) influenced mortality and morbidity at these ages. The results showed that the long gap EA group had more complications and longer hospital stay at age 1 year, and digestive complications were more frequent in the long gap group at age 6 years. The study concluded that long gap strongly influenced digestive morbidity at age 6 years.

In vitro and in vivo evaluation of a pH-, microbiota- and time-based oral delivery platform for colonic release

Summary: A novel drug delivery system based on multiple drug release triggers was developed and evaluated for oral colon delivery, with a specific focus on inflammatory bowel disease (IBD) therapy. The system demonstrated pulsatile drug release in different release media and faster drug release in simulated colonic fluid with fecal bacteria from IBD patients. Promising results were observed in a preliminary rat study, indicating potential therapeutic effects on colitis progression and microbiota alteration.

Summary: This study evaluated the biodistribution and gut toxicity of two commercially available colloidal silver products, Mesosilver (TM) and AgC, in male mice. Both products had similar silver accumulation after high-dose sub-chronic exposure. Physicochemical modifications of silver nanoparticles and ions were observed in the gut. Both products had a moderate impact on the gut barrier at lower doses. The composition of gut microbiota was altered differently by each product. Near-infrared spectroscopy successfully distinguished between the two exposures. This study demonstrates the applicability of biophysics-based methods in understanding the uptake, fate, and toxicological effects of colloidal silver after oral exposure.

Electrospun Filtering Membrane Designed as Component of Self-Decontaminating Protective Masks

Summary: The search for effective protective masks against the 2019 coronavirus outbreak and air pollution has led to the development of advanced filtering textiles with antimicrobial properties. This study successfully produced a filtering electrospun membrane that combines optimal filtration efficiency with fast deactivation of viruses and bacteria. The membrane, composed of polyvinyl alcohol nanofibers stabilized with a biocidal agent, showed promising results against various pathogens, including a human coronavirus strain.

Summary: This study aims to combine the properties of hydrogels based on chitosan (CHT) and the polymer of beta cyclodextrin (PCD) for local delivery of cinnamaldehyde (CN) in diabetic foot ulcers. The results demonstrated the successful development of a cytocompatible (ISO 10993-5) injectable hydrogel with antibacterial and antibiofilm activity. Furthermore, a partial active molecule release and an increase in hydrogel elasticity were observed in the presence of CN.

An extended transcription factor regulatory network controls hepatocyte identity

Summary: Cell identity is controlled by a core transcriptional regulatory circuitry (CoRC) consisting of cell-specific transcription factors (TFs). However, our study reveals that the transcriptional regulatory network regulating hepatocyte identity is more complex than initially thought. We identified a set of TFs, which we call hepatocyte identity (Hep-ID)(CONNECT) TFs, that not only control hepatocyte identity effector genes, but also interact with TFs of the CoRC. Furthermore, we found that Hep-IDCONNECT TFs play a role in fine-tuning CoRC TF expression and can reset CoRC TF expression in dedifferentiated hepatocytes.

Are anxiety and depression associated with cognition and cardiovascular function in young male and female adults?

Summary: Recent studies have found associations between emotional disorders, cognitive impairments, and cardiovascular disorders at the subclinical level. This exploratory study aimed to investigate these relationships in the general population. The results suggest that anxiety and depression are differentially related to cognitive and endothelial functions, but the relationship between cognitive performance and endothelial function was not significant.
