Summary: Chronic stress and alcohol use disorder have a reciprocal relationship and hinder recovery from both disorders. This study identified the TRPV6 channel as being involved in stress, corticosterone, and alcohol-induced intestinal permeability, microbiota dysbiosis, systemic inflammation, and liver damage. TRPV6 deficiency also blocked the effects of stress and stress-mediated potentiation of alcohol-induced dysbiosis of gut microbiota.

Summary: This study investigates the protective effects of betaine on intestinal barrier function in a mouse model of ulcerative colitis and reveals its mechanism of action. The results show that betaine protects the intestinal barrier function of mice with ulcerative colitis by inhibiting the activation of the NF-kappa B-mediated signaling pathway.

Summary: The present study demonstrates that butyrate can alleviate the inflammatory response and tight junction (TJ) injury in bovine mammary epithelial cells (BMECs) by blocking NF-kappa B activation and decreasing inflammatory cytokine production and myosin light chain kinase (MLCK) expression.

Summary: This study revealed the crucial role of GOLPH3 in the development and progression of acute liver and kidney injury induced by endotoxemia. Upregulation of GOLPH3 was associated with high morbidity and mortality. Knockdown of GOLPH3 alleviated cellular stress response and inflammatory reaction, leading to the recovery of Golgi morphology and function. These findings suggest GOLPH3 as a potential therapeutic target for endotoxemia-induced tissue injury.

Summary: The intestinal epithelial tight junction (TJ) barrier plays a crucial role in intestinal diseases, especially inflammatory bowel diseases. The IL-1 beta-induced increase in intestinal TJ permeability is a key factor in the inflammatory process.

Summary: The alpha 9- and alpha 7-containing nicotinic acetylcholine receptors play key roles in physiological and pathological processes. Selective ligands for these receptors are important research tools and potential therapeutic agents. While there is a wealth of information on alpha 7-nAChR ligands, the understanding of selective alpha 9-nAChR ligands is limited. This review provides a comprehensive overview of the latter, along with recent updates on alpha 7-nAChR ligands.

Summary: This study demonstrated the therapeutic potential of CGE against UC by restoring intestinal mucosal barrier integrity, mitigating oxidative stress, inflammatory cascade, as well as inhibiting NF-kappa B/TLR4 and NLRP3 pathways activation in colonic tissues. Additionally, CGE at a dose of 200 mg/kg was more effective in ameliorating DSS-induced UC compared to SAS at the same dose.

Summary: This study aimed to investigate the effects and mechanisms of iE-DAP-induced inflammatory response on the integrity of tight junctions in bovine mammary epithelial cells (BMECs). The results showed that iE-DAP induced an inflammatory response and disrupted tight junctions, which was associated with increased expression of inflammatory cytokines and decreased expression of ZO-1 and Occludin. The inhibition of MLCK and NF-κB signaling pathways reversed the TJ disruption.

Summary: This study investigated the beneficial effect of Lactobacillus paracasei, isolated from koumiss, on a mouse model of diarrhea induced by Escherichia coli O-8 (E. coli O-8). The results showed that L. paracasei reduced intestinal permeability, induced the expression of mucin 2, and increased the number of goblet cells by upregulating the expression of tight junction (TJ) proteins via the nuclear factor kappa B cells-myosin light-chain kinase signaling pathway.

Summary: Thrombocytopenia is common in critical illness, with 18% of Sepsis-3 patients affected. Platelet recovery was more frequent and occurred earlier in survivors, and was not observed until clinical improvement. Thrombocytopenia in Sepsis-3 demonstrated increased disease severity, and patients with platelet count <50 x 10(9)/L showed increased 28 days mortality.

Summary: Survivors of critical illness from COVID-19 may experience persistent long-term impairments in lung function, although most of the functions are restored to normal by 12 months. These patients still have detectable levels of neutralising antibodies against SARS-CoV-2 and all types of IgG at 12 months, but the antibody levels have declined over time.
