Summary: In this study, we investigated whether stem and progenitor cell types have distinct metabolic profiles in the intestinal lineage using the genetically accessible adult Drosophila melanogaster intestine and ex vivo single cell sequencing data. We found that the microRNA miR-277 regulates fatty acid beta-oxidation in the intestinal lineage and can affect the size and survival of stem cells. These findings are important for understanding metabolic differences and dependencies, as well as for identifying potential targets for combinatorial therapy in the treatment of colorectal cancer.

Summary: It has been discovered that PPAR-gamma drives activation-induced glycolysis and promotes IL-9 expression, but not IL-13, in an mTORC1-dependent manner in human T(H)9 cells. The PPAR-gamma-mTORC1-IL-9 pathway is found to be active in T(H)9 cells in human skin inflammation. Furthermore, paracrine IL-9 induces the expression of the lactate transporter, MCT1, in T-H cells and promotes their aerobic glycolysis and proliferative capacity.

Summary: One characteristic of aging is a decline in cell responsiveness to external stimuli, particularly in pathways controlled by hormones. This decline is associated with reduced receptor response to ligands, and it has been observed for hormones such as insulin, leptin, thyroid hormones, and glucocorticoids. This age-related resistance is also seen in adrenergic receptors, adenosine A2B receptor, and other G-protein-coupled receptors. The loss of receptors and changes in lipid composition of neuronal plasma membrane are proposed mechanisms for the decreased sensitivity to hormones and neurotransmitters. The altered membrane properties may contribute to physiological decline, decreased brain function, and increased vulnerability to neuropathology in aging.
