The human papillomavirus family and its role in carcinogenesis

Human papillomaviruses (HPVs) are a family of small double-stranded DNA viruses that have a tropism for the epithelia of the genital and upper respiratory tracts and for the skin. Approximately 150 HPV types have been discovered so far, which are classified into several genera based on their DNA sequence. Approximately 15 high-risk mucosal HPV types are clearly associated with cervical cancer; HPV16 and HPV18 are the most carcinogenic since they are responsible for approximately 50% and 20% of all cervical cancers worldwide, respectively. It is now also clear that these viruses are linked to a subset of other genital cancers, as well as head and neck cancers. Due to their high level of carcinogenic activity, HPV16 and HPV18 are the most studied HPV types so far. Biological studies have highlighted the key roles in cellular transformation of the products of two viral early genes, E6 and E7. Many of the mechanisms of E6 and E7 in subverting the regulation of fundamental cellular events have been fully characterized, contributing not only to our knowledge of how the oncogenic viruses promote cancer development but also to our understanding of basic cell biology. Despite HPV research resulting in extraordinary achievements in the last four decades, significantly improving the screening and prophylaxis of HPV-induced lesions, additional research is necessary to characterize the biology and epidemiology of the vast number of HPV types that have been poorly investigated so far, with a final aim of clarifying their potential roles in other human diseases. Summary: Overall, our study found that alpha HPV types were more prevalent in MSM, beta HPV types were more common in MSW, and gamma HPV types were equally distributed among the three groups. Additionally, several putative novel PV types were identified in the anal mucosa of men with different sexual behaviors and HIV status.

Self-collected and clinician-collected anal swabs show modest agreement for HPV genotyping

Summary: This study found moderate agreement between self-collected anal swabs and clinician-collected anal swabs in HPV genotyping, with no significant difference in the proportion of genotypes detected by either method. Self-collected anal swabs may be used as an alternative to clinician-collected anal swabs for HPV genotyping.

Summary: The study shows that the prevalence of HPV infection varies across anatomical sites in HNC patients in Belgium, with HPV16 being the most common genotype. The proportion of HPV-driven HNC is 10 times higher in OPC compared to OCC and LC in Belgium from 1980 to 2014.

Summary: The study found that beta-HPV detection in skin swabs is a significant predictor of cutaneous squamous cell carcinoma (cuSCC) risk, but only a small subset of cuSCC is etiologically linked to beta-HPV infection. No clear patterns were observed between baseline beta-HPV detection and subsequent development of basal cell carcinoma (BCC), or between baseline gamma-HPV detection and either cuSCC or BCC.

MinION nanopore sequencing and assembly of a complete human papillomavirus genome

Summary: The use of the MinION nanopore sequencer enables the generation of ultra-long reads for more effective characterization of viral genome sequences. The study results demonstrate that the MinION sequencer is an effective strategy for whole-genome sequencing of HPVs with minimal error rates.

Summary: This study in India compared the efficacy of single dose HPV vaccine with two and three doses in protecting against persistent HPV 16 and 18 infection over a 10-year period. The research found that a single dose of the vaccine provided similar protection against these infections as two or three doses, which are responsible for nearly 70% of cervical cancers.

Summary: The study showed that in Rwanda and Bhutan, the school-based national HPV vaccination programs implemented in 2011 and 2010 were highly effective in reducing the prevalence of vaccine-targeted HPV types and other alpha-9 types. The overall effectiveness varied slightly between the two countries, but both demonstrated significant reductions in HPV infection rates.
