Failure to replicate the association between NRG1 and schizophrenia using Japanese large sample

Systematic linkage disequilibrium (LD) mapping of 8p12-21 in the Icelandic population identified neuregulin 1 (NRG1) as a pdrne candidate gene for schizophrenia. However, results of replication studies have been inconsistent, and no large sample analyses have been reported. Therefore, we designed this study with the aim of assessing this putative association between schizophrenia and NRG1 (especially HAP(ICE) region and exon region) using a gene-based association approach in the Japanese population. This study was a two-stage association analysis with a different panel of samples, in which the significant association found in the first-set screening samples (1126 cases and 1022 controls) was further assessed in the confirmation samples (1262 cases and 1172 controls, and 166 trio samples). In the first-set scan, 60 SNPs (49 tagging SNPs from HapMap database, four SNPs from other papers, and seven SNPs detected in the mutation scan) were examined. One haplotype showed a significant association in the first-set screening samples (Global P-value= 0.0244, uncorrected). However, we could not replicate this association in the following independent confirmation samples. Moreover, we could not find sufficient evidence for association of the haplotype identified as being significant in the first-set samples by imputing ungenotyped SNPs from HapMap database. These results indicate that the positionally and functionally attractive regions of NRG1 are unlikely to contribute to susceptibility to schizophrenia in the Japanese population. Moreover, the nature of our results support that two-stage analysis with large sample size is appropriate to examine the susceptibility genes for common diseases. (C) 2008 Elsevier B.V. Summary: This study aimed to examine the factor structure of drug-induced extrapyramidal symptoms in patients with schizophrenia using the Drug-Induced Extrapyramidal Symptoms Scale (DIEPSS). The results identified three factors: F1 (gait and bradykinesia), F2 (muscle rigidity and tremor), and F3 (sialorrhea, akathisia, dystonia, and dyskinesia). These findings suggest that the DIEPSS items could be composed of three different mechanisms.

Positron emission tomography studies in adult patients with attention-deficit/hyperactivity disorder

Summary: ADHD is a neurodevelopmental disorder characterized by inattention, hyperactivity, impulsivity, and cognitive dysfunction. PET studies have not identified characteristic findings for diagnosing ADHD, although disturbances in dopamine, serotonin, and norepinephrine functions have been implicated. Previous PET studies have shown altered binding of dopamine markers in the basal ganglia, but no consistent results were observed for dopamine transporters and receptors. PET studies with ligands related to serotonin and norepinephrine pathways have shown unclear clinical significance or low replicability. Further research is needed to determine characteristic findings for diagnosing ADHD, particularly with new active ligands.

Poor prognostic impact of delirium: especially on mortality and institutionalisation

However, the understanding of delirium remains limited and effective treatments are lacking. This review investigates the effects of delirium on mortality, institutionalization, and dementia in different situations, aiming to clarify the seriousness of its prognosis and provide important guidance for clinical practice and future research.

Serum cortisol and insulin-like growth factor 1 levels in major depressive disorder and schizophrenia

Summary: The pathophysiology of major depressive disorder (MDD) and schizophrenia is related to changes in cortisol and insulin-like growth factor 1 (IGF-1) levels. This study investigated the differences in serum cortisol and IGF-1 levels between patients with MDD, schizophrenia, and healthy controls. The results showed that cortisol levels were higher in schizophrenia patients compared to MDD patients and controls, while MDD patients had higher cortisol levels compared to controls. Both patient groups had higher IGF-1 levels compared to controls, with no significant difference between MDD and schizophrenia patients. These findings suggest pathophysiological differences between these disorders.

A case of schizophrenia with relapsed catatonia successfully treated with blonanserin transdermal patch

Summary: This report describes a 34-year-old female patient with schizophrenia who experienced a relapse of catatonic symptoms after self-discontinuation of prescribed antipsychotic medications. The patient responded well to the transdermal blonanserin patch, which was globally first approved in Japan. This case highlights the potential heterogeneity of catatonia and suggests that antipsychotic blonanserin may be an effective treatment option.

Summary: Autism spectrum disorder (ASD) is caused by a combination of genetic and environmental factors. By analyzing 405 ASD patients, researchers identified disease-causing single-nucleotide variants (SNVs), small insertions and deletions (indels), and copy number variations (CNVs) for molecular diagnoses. They achieved a molecular diagnosis in 16.3% of the patients and found a higher diagnostic rate in females and in simplex cases compared to multiplex families.

Astrotactin 2 (ASTN2) regulates emotional and cognitive functions by affecting neuronal morphogenesis and monoaminergic systems

Summary: Astrotactin2 (ASTN2) regulates neuronal migration and synaptic strength through the trafficking and degradation of surface proteins. Deletion of ASTN2 has been found in patients with schizophrenia, bipolar disorder, and autism spectrum disorder. ASTN2-deficient mice showed increased exploratory activity and social behavior, or decreased despair-, anxiety-like behaviors. These behavioral abnormalities were partially attenuated by risperidone, but not by haloperidol. Dopamine contents were decreased in the striatum, and serotonin or dopamine turnover were increased in multiple brain regions of Astn2 KO mice. Morphological analyses revealed thinning of neural cell layers in the hippocampus, reduction of neural cell bodies in the prefrontal cortex, and decrease in spine density and PSD95 protein in both tissues. ASTN2 deficiency may contribute to the pathogenic mechanism and symptom onset of mental disorders.

Clinical characterization of patients with schizophrenia and 16p13.11 duplication: A case series

Summary: This article reports detailed clinical cases of four patients with schizophrenia and 16p13.11 duplication, with one patient having treatment-resistant schizophrenia and an additional pathogenic rare CNV. Two of the four patients had potential environmental risk factors. The results suggest that a genetic cohort study would be useful in understanding the variable expressivity of 16p13.11 duplication and elucidating the pathophysiology of schizophrenia.

Summary: A polygenic score (Li-PGS(+)) for lithium treatment response in bipolar disorder patients was developed in this study. Li-PGS(+) was found to be positively associated with lithium treatment response. Gene-based analysis also identified 36 candidate genes related to lithium's molecular mechanism.

Summary: This article discusses the difficulties in differential diagnosis in practical psychiatric settings and the prospect of creating a novel diagnostic system using objective biomarkers. Neuroimaging studies have shown that subcortical brain structures are associated with various psycho-behavioral functions, but there are currently no neuroimaging data-driven clinical criteria that can overcome the limitations of the current diagnostic system in reflecting cognitive/social functioning. The authors conducted a large-scale study and found volumetric and lateralization alterations in several psychiatric disorders, which were associated with cognitive/social functioning.

High care and low overprotection from both paternal and maternal parents predict a secure attachment style with a partner among perinatal Japanese women

Summary: This study aimed to examine how paternal and maternal parenting before adolescence affects adult attachment to a partner during the perinatal period using three different models of attachment. The results showed that both paternal and maternal care and overprotection significantly influenced adult attachment, and the four-category and two-category models best described attachment styles.

The three-factor structure of the Autism-Spectrum Quotient Japanese version in pregnant women

Summary: This study aimed to clarify the optimal factor structure of the AQ for assessing autistic traits among perinatal women in Japan. Through exploratory and confirmatory factor analysis on a sample of Japanese women, a model consisting of 25 items distributed across three factors was identified. This three-factor model was found to be superior to previous models in assessing autistic traits.
