Variable inflammation and intramuscular STAT3 phosphorylation and myeloperoxidase levels after downhill running

Individual responses in creatine kinase (CK) release after eccentric exercise are divergent. This study aimed to identify whether this could be related to selected humoral or intramuscular inflammatory factors. Twenty-three subjects were divided into non-exercising (n=5) and downhill run (DHR; n=18) groups (12x5min, 10% decline at 15km/h). Blood samples were analyzed for white blood cell differential count, CK, myoglobin, tumor necrosis factor-, granulocyte colony-stimulating factor, interleukin (IL)-1, IL-6, and IL-10. Muscle biopsies were analyzed for signal transducer and activator of transcription-3 (STAT3), IB, and myeloperoxidase (MPO). DHR participants clustered as early (DHR1) recovery, biphasic response (DHR2), or classic delayed exaggerated CK response (DHR3), with a delayed CK peak (4784 +/- 1496U/L) on day 4. For DHR1 and DHR2, CK peaked on day 1 (DHR1: 1198 +/- 837U/L) or on day 1 and day 4 (DHR2: 1583 +/- 448U/L; 1878 +/- 427U/L), respectively. Immediately post-DHR, IL-6 increased in DHR2 and DHR3 whereas IL-10 increased in all DHR groups. STAT3 signaling increased for DHR1 and DHR2 at 4h, but MPO at day 2 only in DHR2. Objective cluster analysis uncovered a group of subjects with a characteristic biphasic CK release after DHR. The second elevation was related to their early cytokine response. The results provide evidence that early responses following eccentric exercise are indicative of later variation.

Analysis of the regenerative capacity of human serum exosomes after a simple multistep separation from lipoproteins

Summary: This study aimed to develop a multi-step purification protocol to isolate serum exosomes and evaluate their regenerative potential. The purification process of UC DG SEC significantly improved the removal of lipoproteins from serum exosomes, leading to enhanced cellular proliferation and potential angiogenic response in vivo. The purified exosomes showed increased levels of exosome-related proteins and reduced amounts of lipoproteins, indicating a more accurate identification of serum exosome functional activity and composition.

Establishment of fibroblast and myofibroblast phenotypes for use in in vitro co-culture models

Summary: Fibroblasts play a crucial role in wound healing by migrating to the injury site and differentiating into myofibroblasts. Impaired wound healing is associated with a lack of myofibroblasts, resulting in reduced tissue strength and function. Communication between fibroblasts and myofibroblasts, as well as other cellular mediators of repair, influences the outcome of wound repair. However, establishing an in vitro model to investigate this communication is challenging. This study presents a model that allows the retention and culture of fibroblast and myofibroblast phenotypes, and demonstrates the importance of considering paracrine communication during wound healing.

However, under pathological conditions, there can be persistent inflammation without infection, known as sterile metabolic inflammation or metainflammation. This disruption of the immune response is associated with disease progression, immune senescence, and should not be overlooked in clinical management.

Prevalence and aetiology of thyrotoxicosis in patients with hyperemesis gravidarum presenting to a tertiary hospital in Cape Town, South Africa
