Objective. Crohn's disease (CD) and ulcerative colitis (UC) are characterized by an impaired mucosal defence to normal constituents of the intestinal flora and a dysregulated inflammatory response. The purpose of the study was to investigate whether single nucleotide polymorphisms (SNPs) in genes involved in these processes were associated with CD and UC. Material and methods. Allele frequencies of the cyclooxygenase 2 (COX-2/PTGS2/PGHS2) G-765C and breast cancer resistance protein (BCRP/ABCG2) C421A as well as allele and haplotype frequencies of multidrug resistance 1 (MDR1, ABCB1) SNPs G2677T/A, C3435T and G-rs3789243- A (intron 3) were assessed in a Danish case-control study comprising 373 CD and 541 UC patients and 796 healthy controls. Results. Carriers of the homozygous COX-2 and MDR1 intron 3 variant had a relatively high risk of CD, odds ratio (95% CI) (OR (95% CI)) = 2.86 ((1.34-5.88) p = 0.006) and 1.39 ((0.99-1.92) p = 0.054), respectively, and for UC of 2.63 ((1.33-5.26) p = 0.005) and 1.28 ((0.96-1.51) p = 0.093), respectively, assuming complete dominance. No association was found for BCRP or other MDR1 SNPs, or for selected MDR1 haplotypes. No effect-modification of smoking habit at the time of diagnosis was found. Conclusions. An effect of the COX-2 polymorphism on both CD and UC was shown which is compatible with the presence of a recessive allele in linkage equilibrium with the SNP marker in the COX-2 gene. The polymorphism located in intron 3 of the MDR1 gene showed a weak association with CD, and a marginally suggestive association with UC.

Summary: This study examined the association between dietary iron intake and colorectal cancer (CRC) risk. The results showed that in men, higher intake of non-heme iron was associated with a lower risk of CRC, while there was no significant association with total or heme iron intake. In women, there was no significant association between iron intake and CRC risk. Substituting heme iron with non-heme iron was associated with a lower risk of CRC in men.

Summary: By conducting a meta-analysis on the germline genetic data of 234 autophagy-related genes from three independent study populations, we investigated the influence of autophagy-related variants on the risk of Multiple Myeloma (MM) and examined the functional mechanisms behind the observed associations. We identified SNPs within the six CD46, IKBKE, PARK2, ULK4, ATG5, and CDKN2A loci associated with MM risk and found that their effect on disease risk was mediated by specific subsets of immune cells, as well as vitamin D3-, MCP-2-, and IL20-dependent mechanisms.

Summary: This study provides the largest investigation of inflammatory bowel diseases (IBDs) in individuals of East Asian ancestries, identifying 80 IBD loci, including 81 new loci. The study also discovers new IBD genes, such as ADAP1 and GIT2, and improves the accuracy of predicting IBD risk by incorporating data from both East Asian and European ancestries.

Summary: This study examined the promoter hypermethylation of secreted frizzled-related protein 1 (phSFRP1) in cfDNA as a prognostic biomarker and predictor of treatment effect in patients with metastatic FOLFIRINOX-treated PDAC and locally advanced PDAC. The results showed that phSFRP1 is a clinically useful prognostic biomarker in metastatic PDAC and possibly in locally advanced PDAC.

Survival after cancer-related venous thrombosis: the Scandinavian Thrombosis and Cancer Study

Summary: Patients with cancer have a higher risk of developing venous thromboembolism (VTE), which leads to poorer survival compared to cancer alone. This study aimed to investigate the impact of VTE on cancer patients' survival in a general population. The findings showed that cancer patients with VTE had a 3.4-fold higher risk of death compared to those without VTE, regardless of cancer type.

Summary: Inflammatory bowel diseases (IBD) are challenging to manage due to the diversity between patients and the lack of reliable biomarkers. Diet, gut microbiota, genetics, and patient factors are all crucial for disease occurrence and progression. Recent research has identified potential biomarkers for IBD management, but further evaluation is needed. This review provides an update on cutting-edge research in IBD to improve disease management and patient quality of life.

Exploring I-FABP, endothelin-1 and L-lactate as biomarkers of acute intestinal necrosis: a case-control study

Summary: This study aimed to assess intestinal fatty acid binding protein (I-FABP) and endothelin-1 as diagnostic biomarkers for acute intestinal necrosis (AIN). The study found that endothelin-1 alone had a good diagnostic performance, but the diagnostic performance of I-FABP and endothelin-1 combined was only moderate compared to the commonly used biomarker L-lactate.
