Liver X receptor agonist prevents the evolution of collagen-induced arthritis in mice

Objective. Liver X receptors (LXRs) have been characterized as regulators of macrophage inflammatory pathways. Synthetic LXR agonists inhibit the macrophage response to bacterial pathogens and antagonize the induction of a number of pro-inflammatory genes. The aim of this study was to investigate the preventive effects of synthetic LXR agonist, GW3965, treatment on the evolution of arthritis and inflammatory response in a murine CIA model. Methods. Intradermal injection of bovine type II CIA in DBA/1 mice. Along with the induction of CIA, mice were treated with oral GW3965 (0.1, 0.3 or 1.0 mg/kg/day) or vehicle from Day 1 to Day 40. Clinical assessment for arthritis scores and histopathological assessment of joint sections were performed. The expression of inflammatory mediators was evaluated by immunohistochemical staining. Serum pro-inflammatory cytokine levels were determined using ELISA. Results. The CIA incidence was 100% on Day 27 and the severity progressed until Day 35 with histological features of cartilage erosion in vehicle-treated mice. GW3965 treatment significantly reduced the arthritis incidence and attenuated the clinical and histological severity, compared with vehicle-treated mice. GW3965 treatment also significantly reduced inflammatory mediator production in joint sections and serum pro-inflammatory cytokine levels in a dose-dependent manner. Conclusions. These results indicate that activation of LXRs suppresses the onset of CIA and reduces inflammation and joint destruction in CIA mice. The data could suggest that LXR treatment is an effective prophylactic approach to suppress the evolution of synovitis and resultant joint destruction observed in RA.

Effect of statins on the prevention of recurrent thrombosis in thrombotic antiphospholipid syndrome

Summary: This study aimed to assess the effect of statins on the prevention of recurrent thrombosis in patients with thrombotic APS. BASDAI cut-off values corresponding to ASDAS cut-off values

Summary: This study determined the cut-off values of BASDAI for determining disease activity states in ankylosing spondylitis patients. The study found that BASDAI values of 1.9, 3.5, and 4.9 corresponded to ASDAS-CRP values of 1.3, 2.1, and 3.5, respectively. The agreement between disease activity states based on BASDAI and ASDAS-CRP cut-off values was good.

Distinct Gut Microbiota in Patients with Asymptomatic Hyperuricemia: A Potential Protector against Gout Development

Summary: This study reveals differences in microbiota composition between patients with asymptomatic hyperuricemia (asHU) and those with gout. The gut microbiota in asHU patients may have a protective effect against gout development, while in gout patients it may play a role in gout provocation. Uric acid-lowering therapy (ULT) alters the gut microbiota in gout patients, potentially alleviating gout pathology and mitigating gout progression.

Selection of X-ray versus magnetic resonance imaging as a first-line imaging modality for diagnosing axial spondyloarthritis

Summary: This study determines the cut-off values for age and symptom duration to differentiate radiographic axSpA from non-radiographic axSpA, aiming to select the preferred first-line imaging modality. Patients older than 33.5 years with a symptom duration longer than 4.1 years should undergo X-ray imaging as the first-line modality, while patients younger than 33.5 years with a symptom duration less than 4.1 years should undergo MRI as the first-line modality.

Correlation between serologic parameters and disease activity of IgG4-related disease: Differences between patients with normal and elevated serum IgG4 concentrations

Summary: The study aimed to identify serologic parameters associated with the disease activity of IgG4-related disease. In patients with normal serum IgG4 concentrations, only IgG concentration was correlated with disease activity. However, in patients with elevated serum IgG4 concentrations, ESR, C3, C4, IgG4 concentration, IgG concentration, and IgG4/IgG ratio were correlated with disease activity. Among these parameters, the combination of C3 and IgG4 concentration had the strongest correlation coefficient in this group.

Risk of systemic lupus erythematosus flares according to autoantibody positivity at the time of diagnosis

Interleukin-32 as a biomarker in rheumatic diseases: A narrative review

Summary: Interleukin-32 (IL-32) is an important cytokine involved in innate and adaptive immune responses, and its role has been studied in various rheumatic diseases. IL-32 plays different roles in different types of rheumatic diseases and could serve as a biomarker for disease activity or specific manifestations. This narrative review summarizes the associations between IL-32 and various rheumatic diseases and discusses its putative role as a biomarker in each disease.

Risk of all-cause and cause-specific mortality associated with immune-mediated inflammatory diseases in Korea

The results showed that the existence of gut, joint, and skin IMIDs was associated with a higher risk of depression and anxiety. Patients with multiple IMIDs had a higher risk than those with only one.
