Summary: Mutations disrupting the renewal processes of intestinal epithelial cells may contribute to tumorigenesis. Activation of Notch signaling and ablation of p53 induce a highly proliferative and regenerative cell state in intestinal organoids. Yap and Mll1 are elevated in patient-derived colorectal cancer organoids, controlling growth and viability.

In this study, two CDK inhibitors, CYC065 and THZ1, showed promising therapeutic efficacy in GBM patient-derived cell lines, suppressing invasion and inducing cell death. These novel CKIs have the potential to be further investigated for clinical applications in the treatment of GBM.

Summary: This study found that the expression of IRG1 is upregulated in the process of hepatocarcinogenesis, and it promotes hepatocyte apoptosis and liver damage, thus enhancing the development of hepatocellular carcinoma. Mechanistically, mitochondrial IRG1 can associate with anti-apoptotic MCL-1, promoting the activation of Bim and translocation of Bax in the mitochondria, and initiating apoptosis. These findings provide a potential target for preventing liver injury and hepatocellular carcinoma.
