Global gene expression in pseudomyxoma peritonei, with parallel development of two immortalized cell lines

Pseudomyxoma peritonei (PMP) is a rare tumor of appendiceal origin. Treatment is major cytoreductive surgery but morbidity is high. PMP is considered chemoresistant; its molecular biology is understudied; and presently, there is no platform for pre-clinical drug testing. Here, we performed exon array analysis from laser micro-dissected PMP tissue and normal colonic epithelia. The array analysis identified 27 up-regulated and 34 down-regulated genes: candidate up-regulated genes included SLC16A4, DSC3, Aldolase B, EPHX4, and ARHGAP24; candidate down-regulated genes were MS4A12, TMIGD1 and Caspase-5. We confirmed differential expression of the candidate genes and their protein products using in-situ hybridization and immuno-histochemistry. In parallel, we established two primary PMP cell lines, N14A and N15A, and immortalized with an SV40 T-antigen lentiviral vector. We cross-checked for expression of the candidate genes (from the array analyses) using qPCR in the cell lines and demonstrated that the gene profiles were distinct from those of colorectal tumor libraries and commonly used colon cell lines. N14A and N15A were responsiveness to mitomycin and oxaliplatin. This study characterizes global gene expression in PMP, and the parallel development of the first immortalized PMP cell lines; fit for pre-clinical testing and PMP oncogene discovery.

Summary: Research shows that weight loss and increased physical activity can effectively reduce the risk of cancer incidence, but there are still gaps in understanding how these factors are related to cancer and their specific impact on cancer risk. More studies on non-pharmacological weight management are urgently needed in order to support cancer risk reduction and control the growing global burden of cancer.

Indications and outcomes for repeat cytoreductive surgery and heated intra-peritoneal chemotherapy in peritoneal surface malignancy

Summary: This study evaluated the clinical significance of co-localisation of c-MET and VEGFR-2, as well as the association between VEGF pathway-related gene polymorphisms and survival outcomes in women with epithelial ovarian cancer receiving bevacizumab treatment. The results showed that high c-MET/VEGFR-2 co-localisation and VEGFR-2 rs2305945 G/G variant were associated with worse survival outcomes.

Summary: Consistent childhood overweight or obesity may increase the rates of adult obesity-related cancer and decrease the rates of breast cancer. Adult-onset T2D confers additional risk for obesity-related cancer, but the effect does not differ across childhood BMI trajectories.

Summary: This study reviewed data from the UK and Ireland Colorectal Peritoneal Metastases Registry to assess the influence of age on the efficacy of cytoreductive surgery (CRS) with hyperthermic intraperitoneal chemotherapy (HIPEC) for colorectal peritoneal metastases. The results indicated that age did not appear to impact morbidity or long-term survival following CRS and HIPEC. Good survival outcomes were observed when complete cytoreduction was achieved.

A Novel Automated Approach to Mutation-Cancer Relation Extraction by Incorporating Heterogeneous Knowledge

Summary: The automatic extraction of relations between gene mutations and cancer entities from cancer literature using text mining is crucial for precision cancer medicine. In this study, we propose a deep learning model that can jointly extract mutations and their associated cancers. By using two different knowledge bases and integrating them using sentence-based and attribute-aware methods, our model outperforms baseline models and achieves high F1 scores on three public datasets.

Latent class trajectory modelling: impact of changes in model specification

Summary: Latent class trajectory models are often used to identify clinically meaningful patient subgroups, but there is uncertainty and over-reliance on certain evaluation criteria in model selection. Small modifications in model specifications can significantly change the model results, and these models are hypotheses generating and should not be directly implemented in clinical practice.

Summary: This study presents the process of collating colorectal cancer data in the UK and explores the potential use of this data source. By utilizing natural language processing and data integration, longitudinal histories of individual patients can be reconstructed for analysis. The results show that algorithms for analyzing routinely collected data have been refined to provide a rich dataset for researching the natural history, treatment variation, and optimal management of colorectal cancer.
