Summary: This review provides an updated overview of novel gene therapies for high-grade gliomas, highlighting evidence from clinical trials, molecular mechanisms, and future perspectives. The study found that viruses are the most commonly used vehicles, while suicide gene therapies and immunomodulatory gene therapies have shown promising results in some trials.

Summary: Rare germline variants in the WNK2 gene were identified through functional evaluation of a candidate gene and replication in additional SPS cohorts. These variants may affect protein function in the MAPK pathway, thereby increasing the risk for SPS.

Summary: This study aimed to investigate the expression and prognostic value of five microsomal signal peptidase complex subunit genes in head and neck squamous cell carcinoma. The results showed that only SEC11A expression was independently associated with poorer progression-free survival and disease-specific survival. SEC11A upregulation was found in all subtypes of cells in the tumor microenvironment and correlated with the infiltration of certain types of immune cells.

Summary: This study reveals the association between DNA methylation and the silencing of the LRIG1 gene in basal/triple-negative breast cancer. The global demethylating agent 5-aza-2'-deoxycytidine decreases methylation and increases LRIG1 expression. Targeted demethylation and transcriptional activation using CRISPR/deadCas9 technology significantly enhances LRIG1 expression and reduces cancer cell viability.

Summary: We found that SPIB is a tumor suppressor in colorectal cancer cells, sensitizing them to oxaliplatin and 5-FU. SPIB exerts its anti-colorectal cancer effect by activating the NFkB and JNK signaling pathways through MAP4K1. These findings may provide new molecular markers and therapeutic targets for CRC.

Summary: In this study, it was found that the coordinated upregulation of miR-200c and miR-203 in colon epithelial cells is associated with terminal differentiation and plays a role in controlling the expansion capacity and stem cell properties of epithelial cells by suppressing BMI1 expression.

Summary: The study demonstrated that the replication-associated protein (Rep) and RepA encoded by Tobacco yellow dwarf virus (TYDV) have TGS suppressor activity, leading to decreased accumulation of 24-nt siRNAs, suggesting that Rep and RepA can block the steps before the loading of siRNAs into Argonaute (AGO) proteins. This is the first report of TGS suppressors in the genus Mastrevirus.

Summary: The tumor microenvironment (TME) of head and neck squamous cell carcinomas (HNSCC) is complex and heterogeneous, playing a crucial role in effective cancer therapy. Understanding the interaction between cancer inflammation, immunity, oncogenes, and tumor suppressor genes is essential for developing effective cancer treatments. This study aimed to gain a comprehensive understanding of the transcriptomes of the TME in tonsillar squamous cell carcinoma (TSCC), both associated with human papillomavirus (HPV) and not associated with HPV.

Summary: Deletions of chromosome 17p, especially in TP53 gene, are frequently observed in human cancers and associated with poor outcomes. Previous studies have shown that p53-independent mechanisms are involved in 17p deletions-driven cancers. In this study, we found that altered arachidonate metabolism caused by Alox8 deficiency on chromosome 11B3 contributes to B cell malignancy. Upregulation of the cyclooxygenase pathway and increased levels of prostaglandin E2 were observed in cells with Alox8 loss. Ectopic PGE2 inhibited apoptosis and differentiation of pre-B cells. The upregulation of Cox-2 by Alox8 deficiency was found to be crucial for tumorigenesis, and COX-2 inhibitor was effective in treating tumors with Alox8 or 11B3 loss.

Summary: This study found that HIC2 gene is hypermethylated and expressed at low levels in glioma. Hypermethylation and low expression of HIC2 predict poor prognosis. Further experiments showed that HIC2 inhibits glioma proliferation in vitro and in vivo by interacting with RNF44 and PTPRN2.
