Effect of Polypurine Reverse Hoogsteen Hairpins on Relevant Cancer Target Genes in Different Human Cell Lines

We studied the ability of polypurine reverse Hoogsteen hairpins (PPRHs) to silence a variety of relevant cancer-related genes in several human cell lines. PPRHs are hairpins formed by two antiparallel polypurine strands bound by intramolecular Hoogsteen bonds linked by a pentathymidine loop. These hairpins are able to bind to their target DNA sequence through Watson-Crick bonds producing specific silencing of gene expression. We designed PPRHs against the following genes: BCL2, TOP1, mTOR, MDM2, and MYC and tested them for mRNA levels, cytotoxicity, and apoptosis in prostate, pancreas, colon, and breast cancer cell lines. Even though all PPRHs were effective, the most remarkable results were obtained with those against BCL2 and mammalian target of rapamycin (mTOR) in decreasing cell survival and mRNA levels and increasing apoptosis in prostate, colon, and pancreatic cancer cells. In the case of TOP1, MDM2, and MYC, their corresponding PPRHs produced a strong effect in decreasing cell viability and mRNA levels and increasing apoptosis in breast cancer cells. Thus, we confirm that the PPRH technology is broadly useful to silence the expression of cancer-related genes as demonstrated using target genes involved in metabolism (DHFR), proliferation (mTOR), DNA topology (TOP1), lifespan and senescence (telomerase), apoptosis (survivin, BCL2), transcription factors (MYC), and proto-oncogenes (MDM2).

Targeting KRAS Regulation with PolyPurine Reverse Hoogsteen Oligonucleotides

Summary: This study assessed the ability of an engineered liposomal formulation to fuse and function as a nanocarrier in cells. Physicochemical changes on the cell surface and liposome-cell membrane interactions were observed using atomic force microscopy and confocal microscopy. The results confirmed the potential of the engineered liposomal formulation as a nanocarrier.

Base-Mediated Nitrophenyl Reductive Cyclization for the Synthesis of Hexahydro-2,6-methano-1-benzazocines

Summary: A two-step reaction was developed to synthesize a ring system found in natural products. The reaction was explored with eight substituted nitrobenzenes, achieving yields of up to 87%. One of the benzazocine compounds, 4h, exhibited high cytotoxicity and was active against eight cancer cell lines.

Summary: Clonal hematopoiesis (CH) refers to the clonal expansion of mutated hematopoietic stem cells detectable in the peripheral blood or bone marrow through next generation sequencing. This study showed that patients with osteoarthritis have a high prevalence of CH, which involves genes encoding epigenetic modifiers and DNA damage repair pathway proteins. Importantly, it was found that CH can be confined to specific bone marrow spaces and may be eliminated through surgical excision.

Summary: Therapeutic oligonucleotides targeting the overexpressed ERBB2 gene in positive HER-2 breast tumors showed inhibitory effects in cell viability, mRNA and protein levels. Combination with trastuzumab enhanced the therapeutic effects in vitro and reduced tumor growth in vivo. These findings provide preclinical evidence for the potential use of PPRHs as a therapeutic tool for breast cancer.

Targeting MYC Regulation with Polypurine Reverse Hoogsteen Oligonucleotides

Summary: The oncogene MYC plays multiple key roles in cancer, and targeting its expression or function is a promising therapeutic strategy. In this study, Polypurine Reverse Hoogsteen hairpins (PPRHs) were used to target GC-rich regions, including a G-quadruplex-forming region, in the MYC promoter and intronic regions. Two lead PPRHs were found to have high potential for further development as a novel approach for stabilizing the G4 structure and modulating MYC expression.
