The aim of this study was to investigate the effects of caffeine (20 mg/L) intake on cadmium (15 mg/L) accumulation in the rat blood, testes, epididymis and prostate as well as cadmium-induced changes to the antioxidant defense system of the epididymis. Caffeine reduced the cadmium concentration in all tissues analyzed. Meanwhile, cadmium reduced catalase activity and increased superoxide dismutase (SOD) activity in the epididymis. Caffeine increased SOD activity, catalase and glutathione tissue expression and sustains the cadmium's effect on catalase and GSP-Px activity. No differences in the expression of metallothionein and lipid peroxidation were observed among the different treatments in the epididymis. In conclusion, low doses of cadmium alter the antioxidant enzymatic profile of the epididymis, but not induced oxidative lipid damage. Caffeine intake reduces overall cadmium accumulation in the organism and enhances the levels of antioxidant protein expression in the epididymis, thus exerting a protective effect against this metal. Additionally, 42 miRNAs were differentially expressed between severe/critical patients with COVID-19 and mild patients, with some miRNAs significantly associated with disease severity. If validated in a larger cohort, these miRNAs could potentially serve as biomarkers for COVID-19 diagnosis and severity.

OncoTherad(R) is an immunomodulator of biological response that downregulate RANK/RANKL signaling pathway and PD-1/PD-L1 immune checkpoint in non-muscle invasive bladder cancer

Ianny Brum Reis, Luiz Henrique Soares Tibo, Bianca Ribeiro de Souza, Nelson Duran, Wagner Jose Favaro

Summary: Bladder cancer is commonly treated with TUR and OncoTherad(R) immunotherapy. Previous studies with mice have shown that OncoTherad(R) can effectively reduce tumor progression in NMIBC by activating the non-canonical pathway of Toll-like receptors. Immunohistochemical results demonstrate that OncoTherad(R) reduces RANK and RANKL immunoreactivities, indicating a favorable prognosis. Immunohistochemical and western blotting analyses confirm the modulation of PD-1/PD-L1 immune checkpoint by OncoTherad(R).

Summary: BCG-based intravesical immunotherapy is the gold standard treatment for high-risk non-muscle invasive bladder cancer. However, some high-risk patients experience relapse and do not respond to BCG. Our research group developed OncoTherad (R) nano-immunotherapy, which showed positive outcomes and mechanisms of action in an animal model with NMIBC. OncoTherad (R) nano-immunotherapy was more effective than BCG treatment, reducing the malignancy rate by 70% through modulation of TLR4-mediated interferon and RANK/RANKL signaling pathways.

Biogenic Silver Nanoparticles Capped with Proteins: Timed Knowledge and Perspectives

Nelson Duran, Wagner J. Favaro, Silvana Albores, Thyerre S. Costa, Ljubica Tasic

Summary: Biogenic silver nanoparticles are synthesized through the reduction of silver(I) using biomolecules derived from plant extracts, fungal bioproduction, and bacteria. The exact mechanisms of this oxidation-reduction process are not fully understood, but oxidoreductases, cofactors (NADH, FADH2), and phenolic compounds have been identified as the main species involved in the formation of silver(0) and silver nanoparticles. Biosynthesis leads to stable silver nanoparticle colloids with controlled size, shape, and uniformity, and their synthesis is facilitated by biomolecules that also act as surface capping agents. The specific quantity and types of biomolecules that cap or surround silver nanoparticles are still not well-known, and more research is needed to understand the role of capping proteins in the bioactivity of biogenic silver nanoparticles.
