Biomonitoring equivalents for hexachlorobenzene

Recent efforts worldwide have resulted in a growing database of measured concentrations of chemicals in blood and urine samples taken from the general population. However, few tools exist to assist in the interpretation of the measured values in a health risk context. Biomonitoring equivalents (BEs) are defined as the concentration or range of concentrations of a chemical or its metabolite in a biological medium (blood, urine, or other medium) that is consistent with an existing health-based exposure guideline. This study reviews available health-based exposure guidance values for hexachlorobenzene (HCB) from Health Canada, the United States Environmental Protection Agency (US EPA), the US Agency for Toxic Substances and Disease Registry (ATSDR) and World Health Organization (WHO). HCB liver tissue concentrations in chronic rodent bioassays and information on human elimination rates and tissue distribution of HCB were extrapolated to estimate serum lipid-adjusted HCB concentrations that are consistent with the exposure guidance values for HCB. Estimated serum lipid-adjusted HCB concentrations ranging from 16 to 250 ng/g lipid were consistent with non-cancer-based exposure guidance values from various agencies. Concentrations associated with cancer risk-specific doses at target risk levels of interest were also estimated. These BE values may be used as screening tools for evaluation of population biomonitoring data for HCB in a risk assessment context and can assist in prioritization of the potential need for additional risk assessment efforts for HCB relative to other chemicals. Estimation of toluene exposure in air from BMA (S-benzylmercapturic acid) urinary measures using a reverse dosimetry approach based on physiologically pharmacokinetic modeling

Summary: This study used a reverse dosimetry PBPK modeling approach to estimate toluene atmospheric exposure from urinary measurements of S-benzylmercapturic acid. Results showed that estimating toluene exposure based on spot measurements generated more variability and uncertainty compared to 24-h urine samples. Toluene levels estimated were well below Health Canada's maximum chronic air guidelines, suggesting low health risks.

Comprehensive interpretation of in vitro micronucleus test results for 292 chemicals: from hazard identification to risk assessment application

Summary: Risk assessments are increasingly relying on in vitro assays for genotoxicity testing. This study evaluated 292 chemical datasets using the in vitro micronucleus test (MNvit) for hazard identification. The results identified 157 clastogenic chemicals, 25 negatives, and 110 inconclusive substances. In vitro to in vivo extrapolation (IVIVE) was used to convert concentrations into administered equivalent doses (AEDs), which were generally lower than points of departure (PODs) based on in vivo endpoints. Exposure estimates were utilized to calculate bioactivity exposure ratio (BER) values, identifying chemicals that may require additional testing.

Summary: We conducted a study on rats to assess the effects of dose and co-exposure with other rare earth elements (REEs) on the toxicokinetics of praseodymium (Pr) and cerium (Ce). The study showed that dose and mixture exposure are important factors determining the toxicokinetics of REEs.

Summary: In October 2022, the World Health Organization reevaluated the toxic equivalency factors (TEFs) for chlorinated dioxin-like compounds in a panel convened in Lisbon. This effort utilized an updated database, Bayesian dose response modeling, and meta-analysis to derive Best-Estimate TEFs. Applying these new TEFs may result in lower total toxic equivalents for dioxin-like chemicals.
