Summary: Both sensory neurons and immune cells express TRPV1, which can induce neurogenic inflammation and modify tumor growth and metastasis. TRPV1 is also found in various immune cells, and the effects of TRPV1 agonists and antagonists depend on the cell type(s) activated and/or inhibited. A comprehensive understanding of TRPV1 activity on immune cells and nerve endings is necessary to predict the outcome of therapies targeting TRPV1 channels. This review focuses on the neuro-immune modulation of cancer growth and metastasis caused by TRPV1 activation and discusses the potential use of TRPV1 modulators in cancer therapy.

Summary: The authors have engineered a peptide called s-RhTx as a positive allosteric modulator of the TRPV1 channel. It enhances the activity of TRPV1, allowing for reversible elimination of pain-sensing nerve terminals, providing long-lasting analgesia without affecting mice body temperature.

Summary: Capsaicin, the pungent ingredient in chili peppers, can cause intense burning pain in humans, but it has also been used as an analgesic for chronic pain conditions. The analgesic effects of capsaicin depend on concentration, with low concentrations inducing short-term defunctionalization and high concentrations leading to long-term defunctionalization.

Summary: Chronic pain is common during cancer progression, often complicated by bone pain from metastasis, leading to exploration of new mechanisms such as TRP channels for managing cancer-related pain. Studies have shown the potential of TRPV1 and TRPA1 in cancer pain mechanisms, with TRP agonists/antagonists showing promising results in reducing sensitivity in cancer pain models.

Summary: This study aimed to evaluate the ability of immunohistochemistry and quantitative PCR methods to detect Leishmania infection in skin biopsies with lymphoplasmacytic/histiocytic and/or granulomatous dermatitis diagnoses. The study found that immunohistochemistry was the most discriminatory method for estimating the role of the parasite in skin lesions in cases of moderate/severe inflammation.

Summary: RT-qPCR is a sensitive technique to detect minimal residual disease in neuroblastoma patients, with a panel of multiple mRNA markers increasing sensitivity. Two distinct phenotypes in neuroblastoma can be identified by different mRNA markers, and a multiplex RT-qPCR has been developed to test seven mRNA markers while reducing sample volume. This approach shows comparable sensitivity to single marker RT-qPCR, saving time and resources and facilitating clinical introduction.

Summary: Psoriasis is a chronic inflammatory skin disease influenced by both genetic and environmental factors. Increased levels of free nucleic acids in psoriatic skin are associated with IL-23 expression. The study reveals that free nucleic acids regulate IL-23 mRNA expression in epithelial keratinocytes through TLR3 receptor and specific signaling pathways, contributing to the development of an inflammatory milieu favorable for psoriatic symptoms.

Summary: This research studied the pathology and molecular diagnosis of elephant endotheliotropic herpes virus-haemorrhagic disease (EEHV-HD) among Asian elephants. Out of 76 cases, 20 were positive for EEHV infection. The study concluded that Asian elephants in India are latently infected for EEHV1 and in all the fatal EEHV-HD cases, EEHV1A subtype was the causative agent with characteristic pathomorphological changes in visceral organs.

Summary: ABCG2 is a cell membrane pump encoded by the ABCG2 gene, which can protect cells against compounds initiating and/or intensifying neoplasia and is considered a marker of cancer stem cells. Its prognostic significance in colorectal cancer remains unclear. Using publicly available data, ABCG2 was shown to be underexpressed in colon and rectum adenocarcinomas, and this downregulation could result from the methylation level of some sites of the ABCG2 gene. However, no association was found between ABCG2 expression level and survival of colorectal cancer patients.

Summary: HER2-equivocal cases represent around 15% of breast carcinomas, and 20-40% of them are HER2-amplified. The evaluation of HER2 amplification status in equivocal cases is crucial for patient management. In this study, we investigated the performance of STRAT4, a RT-qPCR platform, compared to the recommended methods for the evaluation of HER2-equivocal cases. The results showed that STRAT4 is not reliable for the assessment of HER2 amplification status in equivocal cases.

Summary: Adiponectin plays a crucial role in skin biology, being greatly affected by nutritional status and obesity. The study showed a negative correlation between adiponectin and ADIPOR2 skin expression and serum levels in obese dogs. This suggests that the adiponectin system in the skin undergoes changes in obesity, impacting the local functionality of tissues, including the skin.

Summary: This study characterized the VTG protein in male fathead minnow mucus compared to plasma and liver mRNA. The results showed that the induction kinetics of VTG protein concentration in mucus and plasma were similar, with a significant increase detected after 2 days of exposure in the mucus-based assay and after 7 days in the plasma-based assay.

Summary: Hot peppers, known as chili or chili pepper, are widely used vegetables and spices. The main pungent component, capsaicin, exerts protective effects on cerebral neurons, reducing the risk of stroke and benefiting neurodegenerative diseases. It also reduces immune oxidative and inflammatory signaling to the brain through sensory fibers.
