Precursor ion scan profiles of acylcarnitines by atmospheric pressure thermal desorption chemical ionization tandem mass spectrometry

The fatty acyl esters of L-carnitine (acylcarnitines) are useful biomarkers for the diagnosis of some inborn errors of metabolism analyzed by liquid chromatography/tandem mass spectrometry. In this study the acylcarnitines were analyzed by atmospheric pressure thermal desorption chemical ionization using a commercial tandem mass spectrometer (APTDCI-MS/MS). The method is based on the precursor ion scan mode determination of underivatized acylcarnitines desorbed from samples by a hot desolvation gas flow and ionized by a corona pin discharge. During desorption/ionization step the temperature induces the degradation of acylcarnitines; nevertheless, the common fragment to. all acylcarnitines [MH-59](+) is useful for analyzing their profile. APTDCI parameters, including angle of collection and incidence, gas flows and temperatures, were optimized for acylcarnitines. The experiments were performed drying 2 mu L of an equimolar mixture of acylcarnitine standards on a glass slide. The specificity was evaluated by comparing product ion spectra and the precursor ion spectra of 85 m/z of acylcarnitines obtained by the APTDCI method and by electrospray ionization flow injection analysis (ESI-FIA). The method was also employed to analyze acylcarnitines extracted from a pathological dried blood spot and a control. The method enables analysis of biological samples and recognition of some acylcarnitines that are diagnostic markers of inherited metabolic diseases. The intrinsic high-throughput analysis of the ambient desorption ionization methods offers a new opportunity either for its potential application in clinical chemistry and for the expanded screening of some inborn errors of metabolism. Summary: Rewiring cellular metabolism to recover oxidative metabolism may be an innovative therapeutic strategy for the treatment of spinal cord injury (SCI). In this study, oral administration of metabolic precursors enhanced spinal cord oxidative metabolism, activated the mTORC1 anabolic pathway, increased mitochondrial mass and ROS defense, effectively preventing neural cell apoptosis. These molecular and histological modifications ultimately led to a significant improvement in motor functions.

The results showed that the vaccine provides sustained neutralizing antibody activity against the Alpha variant, but it is less effective against the Gamma and even less against the Delta variants.

Inducible Nitric Oxide Synthase (iNOS): Why a Different Production in COVID-19 Patients of the Two Waves?

Summary: Profound clinical differences were observed between the first and second waves of COVID-19 in Europe. Nitric oxide (NO) may have a positive impact on patients with SARS-CoV-2 infection. The study found that in the first wave, patients had higher levels of serum iNOS, IL-6, and IL-10, in line with the severity of the disease. However, in the second wave, there was no change in iNOS levels with severity, and patients showed lower levels of iNOS, IL-6, and IL-10 compared to the first wave subgroup, suggesting a less severe outcome. In severely ill patients of the second wave, those treated with steroids or azithromycin before hospitalization had significantly lower iNOS levels, indicating a potential impairment in defense against the virus and suggesting NO-based therapies as a potential treatment in patients with low iNOS levels.

VAMS-Based Blood Capillary Sampling for Mass Spectrometry-Based Human Metabolomics Studies

Summary: Volumetric absorptive microsampling (VAMS) is a minimally invasive sample collection method for blood biomolecules analysis. This study investigated the stability of VAMS samples during short-term storage and compared the metabolome obtained from capillary blood collected from the fingertip to those of plasma and venous blood. The results showed that VAMS samples are stable at room temperature for up to 6 hours and capillary blood provides better coverage of the metabolome compared to plasma.

A Targeted Mass Spectrometry Approach to Identify Peripheral Changes in Metabolic Pathways of Patients with Alzheimer's Disease

Summary: Alzheimer's disease is the most common cause of dementia in the elderly population. It not only affects the brain, but also disturbs the overall metabolism of the body. Analysis of blood samples from patients with AD revealed significant dysregulation of metabolites, including upregulation of glycerophospholipids and ceramide, and downregulation of glutamic acid, other phospholipids, and sphingolipids. Pathway analysis showed that at least five metabolic pathways related to polar compounds were dysregulated in AD patients.

Pharmacometabolomics for the Study of Lipid-Lowering Therapies: Opportunities and Challenges

Summary: Lipid-lowering therapies are widely used to prevent ASCVD and related mortality. Omics technologies have been employed to study the mechanisms and effects of these drugs, aiming to improve treatment efficacy and safety. Pharmacometabolomics focuses on studying the effects of drugs on metabolic pathways and the variation of treatment response. This review summarizes significant metabolomic studies on lipid-lowering therapies, including statins, fibrates, and novel drugs and nutraceutical approaches. Integrating pharmacometabolomics data with other omics approaches can help understand the biological mechanisms of lipid-lowering drugs and improve treatment efficacy while reducing side effects.

Mesenchymal stromal cells loaded with paclitaxel show stronger inhibitory effects on survival, migration, and tumorigenicity in TNBC cell lines compared to free paclitaxel, suggesting a potential new drug delivery strategy.
