The aim of the present study was to examine the relevance of depressive symptoms during an acute schizophrenic episode for the prediction of treatment response. Two hundred inpatients who fulfilled DSM-IV criteria for schizophrenia or schizophreniform disorders were assessed at hospital admission and after 6 weeks of inpatient treatment using the Positive and Negative Syndrome Scale (PANSS) and the Hamilton Rating Scale for Depression (HAM-D). Depressive symptoms showed positive correlations with both positive and negative symptoms at admission and after 6 weeks, and decreased during 6 weeks of treatment. Pronounced depressive symptoms (HAM-D scored >= 16) were found in 28% of the sample at admission and in 9% after 6 weeks of treatment. Depressive symptoms at admission predicted a greater improvement of positive and negative symptoms over 6 weeks of treatment, but also more, rather than fewer remaining symptoms after 6 weeks. Both results, however, lost statistical significance when analyses were controlled for the influence of positive and negative symptoms at admission. Therefore, the hypothesis that depressive symptoms are predictive of a favorable treatment response was not supported by the present study. (C) 2007 Elsevier Ireland Ltd. All rights reserved.

Clozapine modulates retinoid homeostasis in human brain and normalizes serum retinoic acid deficit in patients with schizophrenia

Francesca Regen, Nicoleta-Carmen Cosma, Lisa R. Otto, Vera Clemens, Lana Saksone, Janine Gellrich, Berk Uesekes, Thi Minh Tam Ta, Eric Hahn, Michael Dettling, Isabella Heuser, Julian Hellmann-Regen

Summary: Research indicates that the atypical antipsychotic clozapine can inhibit the catabolism of RA, and patients with schizophrenia have lower levels of at-RA and retinol in their serum. However, patients treated with clozapine exhibit higher at-RA serum levels compared to those treated with other antipsychotics.

Using routine MRI data of depressed patients to predict individual responses to electroconvulsive therapy

Matti Gaertner, Elisabetta Ghisu, Ana Lucia Herrera-Melendez, Michael Koslowski, Sabine Aust, Patrick Asbach, Christian Otte, Francesca Regen, Isabella Heuser, Karsten Borgwardt, Simone Grimm, Malek Bajbouj

Summary: ECT is a highly effective treatment for severe and treatment resistant major depression, with 60-80% of patients responding to it. Predicting individual ECT responses using neuroimaging data is promising but faces methodological and practical limitations. Machine learning techniques applied to structural MRI data in this study achieved a 69% accuracy in predicting ECT responses, suggesting potential for overcoming current limitations in translating treatment biomarkers into clinical practice.

Gray matter volume of rostral anterior cingulate cortex predicts rapid antidepressant response to ketamine

Ana Herrera-Melendez, Anna Stippl, Sabine Aust, Milan Scheidegger, Erich Seifritz, Isabella Heuser-Collier, Christian Otte, Malek Bajbouj, Simone Grimm, Matti Gaertner

Summary: This study investigated the impact of ketamine on rapid symptom improvement in depressive patients and found that patients with a larger baseline volume of the bilateral rostral anterior cingulate cortex were more likely to experience rapid symptom reduction. No volumetric changes were observed 24 hours post-treatment.

The VIP-ADHD trial: Does brain arousal have prognostic value for predicting response to psychostimulants in adult ADHD patients?

Maria Strauss, David Petroff, Jue Huang, Christine Ulke, Madlen Paucke, Holger Bogatsch, Pierre Boehme, Knut Hoffmann, Andreas Reif, Sarah Kittel-Schneider, Isabella Heuser, Eike Ahlers, Juergen Gallinat, Daniel Schottle, Andreas Fallgatter, Thomas Ethofer, Stefan Unterecker, Ulrich Hegerl

Summary: EEG studies showed that adult ADHD patients have less stable brain arousal regulation compared to controls, and psychostimulants like methylphenidate have stabilising properties. The study aimed to investigate the predictive value of brain arousal regulation stability in response to methylphenidate therapy in adult ADHD patients. The results indicated that stability in brain arousal regulation may play a role in predicting treatment success with methylphenidate.

Summary: Accumulating evidence suggests the specific involvement of inflammatory processes in major depressive disorder (MDD), particularly affecting innate immunity. Our study found that MDD patients exhibit differences in M1 and M2 polarization under specific cell culture conditions, with gender also playing a significant role in these polarization effects.

Summary: The study investigated the distress patterns in refugees in Berlin and the feasibility of a newly established outpatient clinic. It found that a significant number of refugees from different backgrounds suffered from mental disorders, with depression, PTSD, and adjustment disorder being the most common diagnoses. The findings suggest that a central, low-threshold clinic can effectively provide psychiatric consultations to refugees, helping them overcome barriers in host communities.

Physical and Psychological Consequences of Political Imprisonment in the Soviet Zone of Occupation and the German Democratic Republic

Tolou Maslahati, Aline Lea Voss, Stefan Donth, Isabella Heuser-Collier, Stephan Roepke

Summary: This article provides an overview of the physical and psychological consequences of political imprisonment in the Soviet Zone of Occupation (SBZ) and the German Democratic Republic (GDR), and emphasizes the relevance of the ongoing study at Charite - Universitatsmedizin Berlin. Previous studies suggest an increased prevalence of psychiatric syndromes and physical symptoms in former political prisoners in the SBZ and GDR. However, there is still a need for a systematic assessment of the health effects of political imprisonment in the SBZ/GDR on former prisoners and their offspring.

Summary: This study investigated the presence of aberrant long-range temporal correlations (LRTCs) in the neuronal activities of individuals with attention deficit/hyperactivity disorder (ADHD), and examined the impact of comorbid depression on LRTC and the effectiveness of psychostimulant therapy. The results showed significant LRTCs in all investigated frequency bands, but no significant differences in LRTC between ADHD patients with and without depressive symptoms. However, depressed ADHD patients seemed to benefit more from psychostimulant therapy based on self-report.

Acute and Chronic Macrophage Differentiation Modulates TREM2 in a Personalized Alzheimer's Patient-Derived Assay

Nicoleta-Carmen Cosma, Neriman Eren, Berk Usekes, Susanna Gerike, Isabella Heuser, Oliver Peters, Julian Hellmann-Regen

Summary: Neuroinflammation is crucial in the development of Alzheimer's disease (AD), and the modulation of TREM2 expression in brain macrophages is important in the immune response to AD pathology. In this study, a patient-specific model using cells from AD patients and matched controls was used to assess TREM2 synthesis. The effects of different types of macrophage differentiation and retinoic acid treatment on TREM2 synthesis were evaluated. The results showed that M2 differentiation increased TREM2 synthesis in control cells, while chronic M2 and M0 differentiation increased TREM2 synthesis in both AD and control cells. Interestingly, retinoic acid treatment had no effect on TREM2 expression. This personalized model could be used to screen potential drug-mediated treatment responses in vitro.
