In the recent years, global proteomics approaches have been widely used to characterize a number of tissue proteomes including plasma and liver; however, the elevated complexity of these samples in combination with the high abundance of some specific proteins make the study of the lowest abundant proteins difficult. This review is focused on different strategies that have been developed to extend the proteome focused on these two tissues, as, for example, the analysis of sub-cellular proteomes. In this regard, two special kind of extracellular vesicles exosomes and membrane plasma shedding vesicles are emerging as excellent biological source both to extend the liver and plasma proteomes and to be applied in the discovery of non-invasive liver-specific disease biomarkers.

Summary: This study presents a J-Edited Diffusional (JEDI) proton NMR spectroscopic approach that selectively enhances signals from inflammatory markers in blood serum. The results show that JEDI is an improved method for selectively investigating inflammatory signals in serum and may have widespread diagnostic applicability to disease states associated with systemic inflammation.

Summary: MAT alpha 1 plays a role in reducing activity and causing mitochondrial dysfunction in alcohol-associated liver disease (ALD). This study found that the peptidyl-prolyl cis/trans isomerase PIN1 and kinase CK2 mediate the selective depletion of mitochondrial MAT alpha 1 in ALD. Blocking the interaction between PIN1 and MAT alpha 1 increased mitochondrial levels of MAT alpha 1 and protected against alcohol-induced mitochondrial dysfunction and fat accumulation.

Summary: This study investigates the role of extracellular vesicles (EVs) in modulating the tumor microenvironment and their impact on the development of TSC tumors. It shows that EVs secreted from TSC2-deficient cells contain a specific protein cargo that promotes cell viability, proliferation, and growth factor secretion in the tumor microenvironment. The study also demonstrates that rapamycin can alter the cargo of EVs and reduce their ability to promote cell proliferation.

Orally Administered Bifidobacterium adolescentis Diminishes Serum Glutamate Concentration in Mice

Summary: This study used mice as a model and fed them with a bacterial strain of B. adolescentis that can degrade glutamate and convert it into GABA. The bacterium can survive in the gastric tract and the animals reduce their blood glutamate concentration over time. The results suggest that an oral diet with this probiotic-type bacteria could reduce the concentration of glutamate in the blood, providing a reference for clinical trial studies in patients with chronic diseases.

A Novel Approach on the Use of Samples from Faecal Occult Blood Screening Kits for Metabolomics Analysis: Application in Colorectal Cancer Population

Summary: Colorectal cancer (CRC) is increasing in incidence and is currently the third most common cancer. Non-invasive, specific, and accurate diagnostic tools are needed for early diagnosis and prognosis. This study evaluated the feasibility of using extracts from fecal occult blood (FOB) kits for metabolomics studies, and identified cholesteryl esters with polyunsaturated fatty acids (PUFAs) as relevant metabolites for separating CRC patients from control individuals and adenoma patients. The results highlight the advantage of using the same FOB samples for multiple analyses and suggest potential future perspectives in lipid metabolism.

The Role of Longevity Assurance Homolog 2/Ceramide Synthase 2 in Bladder Cancer

Summary: The human CERS2 gene encodes a protein called CERS2, which has been found to have antitumor effects in various types of cancer, but promotes tumor growth in adenocarcinoma. This review focuses on the influence of CERS2 in bladder cancer, including its structure, activity, and the miRNAs that regulate its expression. Mechanistically, CERS2 may act as an antitumor protein through the production of long-chain ceramides, interaction with vacuolar ATPase, and inhibition of mitochondrial fission. Additionally, the expression of CERS2 in bladder cancer and its association with disease progression are discussed.
