Summary: Pharmaceutical cocrystals and salts are extensively studied for their ability to modify the physicochemical properties of active pharmaceutical ingredients. In this study, different forms of olanzapine were discovered through solvent drop grinding and ball milling. These new solid phases all followed the Delta pKa rule and showed increased solubility due to strong hydrogen bonding interactions.

Summary: The broadly neutralizing antibody CAP256-VRC26.25 has exceptional potency against HIV-1 and several unusual features were observed during its characterization and production. Through altering the site of proteolytic cleavage and adding a half-life extending mutation, the engineered antibody CAP256V2LS retained its neutralization potency, had favorable pharmacokinetics, and was suitable for large scale production.

Summary: In this study, a single-chain variable fragment (scFv) targeting VEGFR-2 was successfully engineered using a CDR grafting technique to enhance solubility and antiangiogenic properties. The grafted scFv molecule showed improved solubility and similar binding affinities to VEGFR-2 compared to the original molecule, while also demonstrating effective antiangiogenic activity in biological assays.

Summary: In this study, two novel solids, crystalline FMT-MT and vitreous FMT-MTa, were successfully obtained using crystal engineering concepts and the co-amorphous formation strategy. The crystalline FMT-MT showed higher physical stability, while the vitreous FMT-MTa exhibited the inherent features of amorphous phases. The solubility assays revealed that both FMT-MT and FMT-MTa were more soluble than the marketed polymorph, with 2.02-fold and 2.68-fold increases in solubility, respectively, in water.

Summary: In this study, a novel fusion protein (L19-IFN gamma KRG) is described, which selectively localizes to tumors and regains biological activity upon antigen binding. This fusion protein showed tumor growth inhibition and increased intratumoral concentration of T cells and NK cells in murine cancer models, with a better effect when used in combination with anti-PD-1 therapy.

The sulfation modification of tyrosine residues further enhances the diversity of the antibody repertoire. This review article provides an overview of the mechanism of tyrosine sulfation modification and its application in antibody engineering and production, while also discussing the challenges and opportunities in this field.

Summary: In this study, an engineered human anti-ETA antibody, MJF1-PFc29, with improved thermostability and ETA binding affinity, was developed. The antibody showed inhibitory effects on tumor cell proliferation and exhibited stronger tumor growth inhibition in a colorectal cancer mouse model compared to MJF1. These findings indicate that MJF1-PFc29 has great therapeutic potential and high antitumor potency against various types of cancers, including colorectal cancer.
