Polysaccharide co-polymerases: the enigmatic conductors of the O-antigen assembly orchestra

The O-antigen lipopolysaccharides on bacterial surface contain variable number of oligosaccharide repeat units with their length having a modal distribution specific to the bacterial strain. The polysaccharide length distribution is controlled by the proteins called polysaccharide co-polymerases (PCPs), which are embedded in the inner membrane in Gram-negative bacteria and form homo oligomers. The 3D structures of periplasmic domains of several PCPs have been determined and provided the first insights into the possible mechanism of polysaccharide length determination mechanism. Here we review the current knowledge of structure and function of these polysaccharide length regulators.

Summary: Lipopolysaccharide (LPS) is a characteristic molecule of the outer leaflet of the Gram-negative bacterial outer membrane, consisting of lipid A, core oligosaccharide, and O antigen. It plays an important role in reducing the permeability of antibiotics, toxic cationic metals, and antimicrobial peptides, and modulates innate immune responses.

Summary: Achromobacter species can cause chronic and potentially severe infections in immunocompromised patients, especially in those with cystic fibrosis. Analysis of Achromobacter clinical isolates reveals that intrinsic resistance to the antimicrobial peptide polymyxin B (PmB) is prevalent, and many isolates show increased resistance upon PmB challenge. The analysis of lipid A modifications in Achromobacter isolates shows that PmB-resistant strains display modifications that can enhance resistance to PmB and potentially other antimicrobial peptides.

Summary: This study utilized the protein O-glycosylation pathway in Burkholderia to develop a diagnostic tool for glanders. The newly developed diagnostic method showed high specificity and sensitivity in detecting glanders-infected horses, providing crucial support for the control and elimination of Burkholderia infections. The results indicate that protein O-glycosylation in Burkholderia can serve as a biomarker for the diagnosis of Burkholderia-associated infections.
