Summary: The bioluminescence of hydromedusan photoproteins, responsible for marine jellyfish and hydroid, is a unique biochemical system consisting of apoprotein and preoxygenated coelenterazine. The role of water molecule and dioxetanone intermediate in the bioluminescence reaction is still under discussion.

Summary: Chemi- and bioluminescence are remarkable light-emitting phenomena that convert thermal energy into excitation energy through (bio)chemical reactions. In this study, a novel dibrominated Coelenterazine analog with superior chemiluminescent properties was synthesized and characterized. This compound exhibited enhanced luminescence in terms of total light output and emission lifetime compared to natural imidazopyrazinones and commercially available analogs in aprotic media, showing potential as a basis for optimized probes for reactive oxygen species.

Summary: Coelenterazine-v (CTZ-v) is a suitable substrate for deep tissue imaging with Renilla luciferase, but Ca2+-regulated photoproteins activated by CTZ-v have low bioluminescence activity and are unsuitable for calcium imaging. The low bioluminescence activity of obelin-v is attributed to the low efficiency of generating an electronic excited state, possibly due to small changes in the substrate microenvironment at the active site.

Summary: Enzyme-based technologies are crucial for environmentally friendly industrial and biotech processes, but a thorough understanding of enzyme-catalytic mechanisms is necessary. This review outlines a strategy using reconstructed ancestral enzymes and unconvertible substrate analogues to decipher challenging enzyme catalysis mechanisms, which is particularly useful for enzymes with unknown catalytic mechanisms and poor crystallizability.

Summary: The present study introduces a unique BL signature imaging system with novel CTZ analogues named C-series. It examines the BL intensity patterns and spectra of nine C-series CTZ analogues according to marine luciferases. The results show that four CTZ analogues, C3, C4, C6, and C7, luminesce with distinctive BL spectral signatures and intensity patterns according to different luciferases. These findings provide a unique methodology to specify each signal in a single- or multi-reporter system using unique BL signatures and patterns of reporter luciferases.

Summary: Cancer is a challenging disease to treat, and this study focuses on the design, synthesis, and evaluation of different brominated coelenteramines as potential anticancer molecules. The results highlight the importance of the 4-bromophenyl moiety for their anticancer activity and suggest independent mechanisms of action compared to coelenterazines. One of the studied coelenteramines showed promise in improving the profile of a known chemotherapeutic agent.

Summary: Halogenated derivatives of Coelenterazine, specifically brominated Coelenteramide and Coelenteramine, have been found to exhibit potent anticancer activity against gastric and lung cancer cells. These compounds show a certain level of safety towards noncancer cells and induce cell death through apoptosis.

Summary: The lantern shark genus Etmopterus consists of approximately 40 species of bioluminescent cartilaginous fishes that emit blue light mainly from their ventral body surface. This study detected both coelenterazine and coelenterazine-dependent luciferase activity in the ventral photophore tissue of Etmopterus molleri, suggesting that bioluminescence in lantern sharks is produced using coelenterazine as a substrate for the luciferin-luciferase reaction, similar to some luminous bony fishes.

Summary: The mode of action of brominated Coelenteramine towards gastric cancer cells was investigated using synchrotron radiation-based FTIR spectroscopy. This method revealed that the anticancer activity of the compound is closely related to cellular lipids, impacting their organization and composition. Increased oxidative stress led to changes in membrane polarity, lipid chain packing, and lipid composition, which were not observed in noncancer cells, explaining its selectivity profile. Therefore, synchrotron radiation-based FTIR proved to be a powerful technique for studying the mechanism of anticancer drugs.

Summary: The bioluminescence of ctenophores is controlled by light-sensitive Ca2+-regulated photoproteins, which have been cloned and characterized from four different species. The specific activity, quantum yield, and absorbance characteristics of recombinant berovin were investigated, with values found to be lower than cnidarian photoproteins. The study provides insight into the differences in amino acid environments between ctenophore and cnidarian photoproteins.

Despite the discovery of over 700 genera of bioluminescent organisms, there is still insufficient knowledge about their emission patterns under natural and stress conditions. Various stimulation techniques, such as chemical, mechanical, photic, thermal, magnetic, and electrical, can be used to identify organisms that have not been recognized as luminescent.
