Summary: Muscle stem cells are crucial for the maintenance and repair of skeletal muscles in mammals. It has been found that a specific type of RNA modification, known as N6-methyladenosine (m6A), plays a significant role in muscle development and regeneration. However, the exact mechanism by which m6A modification regulates muscle stem cells is not well understood. In this study, researchers discovered that a protein called Ythdc1, which is responsible for reading m6A modifications, is essential for skeletal muscle regeneration and the proliferation of muscle stem cells in mice. The absence of Ythdc1 prevents muscle stem cells from exiting a quiescent state, hindering their ability to regenerate muscle tissue. This is due to Ythdc1's role in regulating the alternative splicing of specific mRNAs and, subsequently, the PI4K-Akt-mTOR signaling pathway. Ythdc1-deficient muscle stem cells exhibit deficiencies in key molecules involved in this pathway, leading to their inability to exit quiescence. These findings establish a connection between dynamic RNA methylation and the regulation of the PI4K-Akt-mTOR pathway during stem cell proliferation and adult tissue regeneration.

Summary: Decellularized extracellular matrix scaffold is widely used for organ engineering, but matrix decomposition and inflammation after transplantation hinder its clinical success. In this study, nano-graphene oxide was used to modify the properties of decellularized liver scaffolds, resulting in enhanced resistance to degradation and reduced inflammation. The introduction of nano-graphene oxide also protected the scaffolds against in vivo degradation, leading to functional bioengineered livers and improved therapeutic effects.

Summary: Recent studies have shown that impaired renewal of type 2 alveolar epithelial cells (ATII) in IPF patients and mice with defective ATII can lead to severe pulmonary fibrosis. Growth hormone receptor (GHR) expression, mainly found in mesenchymal cells, is significantly decreased in IPF lungs, but higher levels of GHR expression are associated with better lung function in IPF patients. The findings suggest that a previously unidentified mesenchymal paracrine signaling via GHR can support ATII progenitor cell renewal and limit the severity of lung fibrosis.

Summary: This study identified a deficiency of the zinc transporter SLC39A8 (ZIP8) in Type 2 alveolar epithelial cells (AEC2s) from both idiopathic pulmonary fibrosis (IPF) lungs and lungs of old mice, which is associated with impaired AEC2 renewal capacity and enhanced lung fibrosis. The regulation of AEC2 progenitor function by ZIP8 is dependent on SIRT1. Replenishment with exogenous zinc and activation of SIRT1 promote self-renewal and differentiation of AEC2s, and therapeutic strategies targeting zinc metabolism and SIRT1 signaling could potentially improve AEC2 progenitor function and mitigate fibrogenesis.
