The biosynthesis and metabolism of astaxanthin in coenobium alga Scenedesmus obliquus were investigated using a two-stage culture. The first stage was for the analysis of biosynthesis and accumulation of astaxanthin in alga cells which were cultured under induction conditions (incubation at 30 degrees C and illumination of 180 mu mol m(-2) s(-1)) for 48 h. The composition of the secondary carotenoids in algal cells was analyzed and seven ketocarotenoids were identified. The results implied that S. obliquus synthesized astaxanthin from beta-carotene through three possible pathways. In the second stage, the cultures were transferred to normal conditions (incubation at 25 C and illumination of 80 mu mol m(-2) s(-1)) for 72 h. Algal cells accumulated more chlorophyll and biosynthesis of secondary carotenoids terminated, the content of secondary carotenoids decreased from 59.48 to 6.57%. The results inferred that accumulation and metabolism of astaxanthin could be controlled by cultivated conditions which also could lead the mobilization of secondary carotenoids to support the algal cell growth. The results also implied that presumed conversions from astaxanthin to lutein or antheraxanthin could be modulated by culturing conditions. (C) 2008 Published by Elsevier Ltd.

Summary: This study identified suitable chloroplast molecular markers for phylogenetic analyses of Chaetophorales, indicating that the limited number of samples was not the reason for the unsupported phylogenetic relationships. This research provides insights for solving taxonomic problems of the order Chaetophorales.

Chloroplast genomes of five Oedogonium species: genome structure, phylogenetic analysis and adaptive evolution

Qian Xiong, Yuxin Hu, Wenqi Lv, Qinghua Wang, Guoxiang Liu, Zhengyu Hu

Summary: The study identified five chloroplast genomes of Oedogonium species, revealing their structural conservation but variation in genome sizes, AT contents, introns, and repeats. Phylogenetic analyses suggested that both Oedocladium and Oedogonium are polyphyletic groups. The identification of a positively selected gene in certain species indicates adaptive evolution in terrestrial Oedogoniales taxa.

Chloroplast Genome Traits Correlate With Organismal Complexity and Ecological Traits in Chlorophyta

Jiao Fang, Yangliang Chen, Guoxiang Liu, Heroen Verbruggen, Huan Zhu

Summary: Studies have shown a positive relationship between cell size and chloroplast genome size in chloroplast-bearing protists. The size of chloroplast genomes varies significantly between green algae in freshwater, marine, and terrestrial habitats, with terrestrial algae generally having larger genomes. The length of intergenic regions is a key factor contributing to the larger genomes in terrestrial species. Additionally, there are differences in chloroplast genome size and GC content among different functional morphological categories of algae.

Phyllosphere eukaryotic microalgal communities in rainforests: Drivers and diversity

Ben -Wen Liu, Shu-Yin Li, Huan Zhu, Guo-Xiang Liu

Summary: This study aims to identify the environmental factors that drive phyllosphere algal community composition and diversity in rainforests. The researchers used single molecule real-time sequencing to characterize the phyllosphere microalgal communities growing on four common tree species in three types of forest. The study found that forest type and host tree species significantly influenced the structure of the algal community, and that soluble reactive phosphorous, total nitrogen, and ammonium contents were important environmental factors affecting the communities.
