Pharmacological inhibition of PRMT4/5/7 was shown to suppress cancer cell growth with synergistic effects when co-inhibited.

Summary: In order to understand the function of protein arginine methyltransferases(PRMTs), it is crucial to identify their substrate proteins. However, this is challenging due to the dominance of stable and strong interacting proteins over weak and transient substrate proteins. In this study, a novel photoreactive probe-based strategy was developed to identify the substrate proteins of methyltransferases, specifically PRMT1. This strategy effectively distinguished substrate proteins from other interacting proteins and allowed the identification of highly confident substrate proteins. Importantly, it was discovered that hypomethylation of proteins is a prerequisite for efficient capturing of substrate proteins. This study describes the development of a robust chemical proteomic tool for profiling transient substrates and has broad biomedical applications.

Summary: Histone methylation is an important epigenetic regulatory mechanism in plants, affecting gene expression by altering the methylation status of histone tails. Protein arginine methyltransferases (PRMTs) are responsible for histone methylation of specific residues, important for plant development and stress adaptation. This study identified and characterized eight PRMT genes in maize, with ZmPRMT1-8 categorized into three subfamilies based on phylogenetic analyses. Overexpression of ZmPRMT1 in transgenic Arabidopsis resulted in earlier flowering and enhanced heat tolerance, indicating its role in regulating flowering time and heat stress response. These findings provide a theoretical basis for understanding the functional roles and epigenetic mechanisms of ZmPRMT genes in maize growth, development, and responses to abiotic stresses.

Summary: Approximately 15-25% of acute lymphoblastic leukemias (ALL) originate from T-lineage cells, with T-ALL having a less favorable prognosis compared to B-ALL. High expression of PRMT7 was identified in T-ALL cells, and genetic deletion of PRMT7 decreased colony formation and altered arginine monomethylation patterns in protein complexes associated with RNA and DNA processing. Disruption of arginine monomethylation patterns was observed in proteins linked to T-ALL pathogenesis, such as RUNX1, leading to deregulated target gene expression. These findings suggest that PRMT7 plays a crucial role in T-ALL pathogenesis.
