A growing number of studies have highlighted the role of microRNAs (miRNAs or miRs) in the development and progression of cancer. In particular, the aberrant expression of cancer-related proteins, such as oncogenes and tumor suppressors has been shown to correlate with the modulation of the expression of specific miRNAs. In the present study, we aimed to determine which downregulated miRNAs may be involved in modulating the expression of the oncogenic transcription factor, Yin Yang 1 (YY1). YY1 has been reported to be overexpressed in several malignancies and our previous studies have highlighted the significant correlation between the levels of YY1 and aggressive behavior in non-Hodgkin's lymphoma (NHL). A total of 57 miRNAs that are potentially capable of targeting YY1 was identified through in silico approaches. The search of publicly available NHL datasets, including paired mRNA and miRNA data (GSE23026) highlighted a significant correlation (Pearson's correlation, r> 0.5) between the expression levels of YY1 and the expression levels of a limited set of miRNAs, including miR-363, miR-200a, miR-23b, miR-15a and miR-15b. Intriguingly, both hsa-miR-363 and hsa-miR-200a belong to the top 20 miRNAs that were found to be downregulated in Burkitt's lymphoma (BL) tissue compared to normal tissue. Although further validation studies are warranted, the identification of these two miRNAs associated with the upregulation of YY1 in BL may provide further insight into the pathogenesis of this tumor and may contribute to more personalized and targeted treatment approaches for patients with this disease.

Summary: This study demonstrates the feasibility of using NGS approach for comprehensive molecular profiling of PDAC patients, regardless of their surgical eligibility. The mutational profiles detected in cytologic samples showed high concordance with the corresponding histologic specimens obtained after surgery, and actionable mutations were identified in a significant proportion of patients.

Summary: The study examined whether one-year nutrition counseling in the context of a Mediterranean diet, with or without low-GI carbohydrates counseling, could influence the DII in women with breast cancer. The results showed that adherence to the Mediterranean diet and lower dietary GI independently contributed to the decrease in DII scores.

In Silico Identification of a BRCA1:miR-29:DNMT3 Axis Involved in the Control of Hormone Receptors in BRCA1-Associated Breast Cancers

Manuela Santarosa, Davide Baldazzi, Michela Armellin, Roberta Maestro

Summary: Germline inactivating mutations in the BRCA1 gene increase the lifetime risk of ovarian and breast cancer. The role of miRNAs and their networks in mediating BRCA1 functions and the potential mechanism of how BRCA1 inactivation contributes to the specific phenotype of triple-negative breast cancer (TNBC) were investigated. The miR-29:DNA methyltransferase network was found to control hormone receptor expression through the methylation level of HR genes, suggesting its involvement in the receptor-negative phenotype of BRCA1 dysfunctional tumors.

Summary: This study investigated the role of the PDE5A isoenzyme and epigenetic mechanisms in female genital tissue disorders. The results revealed differences in miRNA expression levels and PDE5A expression levels between women with FGAD and healthy women. These findings suggest that treatment with PDE5 inhibitors may be beneficial for women with FGAD.

Summary: This study found that LGG-SN has anti-proliferative activity against colon cancer and metastatic melanoma cells and enhances the efficacy of anti-cancer drugs. This suggests that LGG could be a new therapeutic approach in oncology to reduce chemotherapy-related side effects and overcome resistance issues, improving the treatment response in cancer patients.

In silico analysis of the solute carrier (SLC) family in cancer indicates a link among DNA methylation, metabolic adaptation, drug response, and immune reactivity

Summary: The oncogenic transformation is driven by genetic and epigenetic alterations, resulting in metabolic reprogramming. This study identified deregulated solute carrier (SLC) transporters in various tumors and investigated their relationship with tumor features and DNA methylation. Key SLCs were found to be associated with tumor development, immune escape, and chemoresistance. These findings provide insights into potential cancer biomarkers and therapeutic targets.

Clinical Relevance of Targeted Therapy and Immune-Checkpoint Inhibition in Lung Cancer

Gian Marco Leone, Saverio Candido, Alessandro Lavoro, Silvia Vivarelli, Giuseppe Gattuso, Daniela Calina, Massimo Libra, Luca Falzone

Summary: Lung cancer is the second most diagnosed tumor with the highest mortality rate. Recent progress in the treatment of lung cancer includes targeted therapies and immunotherapy, which have been approved in clinical practice. This review discusses the current and ongoing clinical studies on targeted therapies and immune-checkpoint inhibitors for lung cancer, as well as the advantages and limitations of these new therapeutic approaches. Furthermore, the importance of human microbiota as biomarkers and therapeutic targets for lung cancer is analyzed. The future research milestones in personalized treatment for lung cancer are expected to consider the genetic landscape, immune background, and individual variables such as patient-specific gut microbial composition.

Summary: The ketogenic diet (KD) has gained attention as a potential therapeutic approach in breast cancer (BC), but its effects on BC remain unclear. Some studies suggest potential benefits of the KD in inhibiting tumor growth and improving treatment response, but robust clinical trials providing clear evidence are lacking. Concerns exist regarding the safety and long-term effects of sustained ketosis in cancer patients. Therefore, caution should be exercised in the application of KD in BC treatment, and evidence-based standard of care treatments should continue to be prioritized.
