Summary: Disruption to the ER-Golgi network can cause neurodevelopmental disorders, and ARF3 mutations have been identified as the cause of a rare pediatric neurological disorder. This study provides detailed molecular characterization of ARF3 mutations and demonstrates their impact on Golgi morphology, vesicles assembly, and trafficking.

Summary: Wound healing is a complex biological process influenced by changes in the microenvironment, with cell polarity being a significant parameter. A method utilizing carbon dots to monitor cell polarity during wound healing has been successfully developed, with dynamic changes in polarity monitored for the first time in zebrafish. This study provides an important experimental basis for investigating wound healing using polarity-sensitive CDs.

Summary: Asthma exacerbation is triggered by airway macrophages through a prion-like cell-to-cell transmission of extracellular particulates. Mice with small GTPase Arf6-deficient macrophages show alleviated allergic asthma and reduced IL-1 beta production. The Arf6-dependent intercellular transmission of extracellular ASC specks contributes to the amplification of allergic inflammation and subsequent asthma exacerbation.

Summary: During HPV entry, the virus is trafficked through the cell compartments, including the endosome, trans-Golgi network (TGN), and Golgi apparatus. The coat protein complex I (COPI) is identified as a host factor required for HPV infection. The interaction between the cytoplasmic segment of HPV L2 and COPI facilitates the trafficking of HPV through the TGN and Golgi stacks, ultimately reaching the nucleus.

Summary: Natural molecules/extracts have beneficial effects on wound healing processes. Polysucrose-based hydrogels have been synthesized with in situ loading of natural molecules/extracts such as Manuka honey, Eucalyptus honey, Ginkgo biloba, thymol, and metformin. The release profiles of these substances from the hydrogels followed a quasi-Fickian diffusion mechanism. In vitro studies showed that a combination of Ginkgo biloba and Eucalyptus honey in the hydrogel scaffold can accelerate tissue regeneration during wound healing.

Summary: We report a pediatric patient with an ARF1-related disorder associated with a missense variant (c.296 G > A; p.R99H) in the ARF1 gene, causing developmental delay, hypotonia, intellectual disability, and motor stereotypies. Neuroimaging revealed brain abnormalities. The R99H-ARF1 variant protein showed alterations in Golgi apparatus, coat proteins recruitment, and endosomal morphology, similar to constitutively active ARF1 mutants. This study suggests the pathogenesis mechanism of the R99H variant involves constitutive activation and highlights the phenotypic variability of ARF1-related disorders.
