Apoptosis signal-regulating kinase 1 and cyclin D1 compose a positive feedback loop contributing to tumor growth in gastric cancer

Mitogen-activated protein kinase (MAPK) pathways regulate multiple cellular functions and are highly active in many types of human cancers. Apoptosis signal-regulating kinase 1 (ASK1) is an upstream MAPK involved in apoptosis, inflammation, and carcinogenesis. This study investigated the role of ASK1 in the development of gastric cancer. In human gastric cancer specimens, we observed increased ASK1 expression, compared to nontumor epithelium. Using a chemically induced murine gastric tumorigenesis model, we observed increased tumor ASK1 expression, and ASK1 knockout mice had both fewer and smaller tumors than wild-type (WT) mice. ASK1 siRNA inhibited cell proliferation through the accumulation of cells in G1 phase of the cell cycle, and reduced cyclin D1 expression in gastric cancer cells, whereas these effects were uncommon in other cancer cells. ASK1 overexpression induced the transcription of cyclin D1, through AP-1 activation, and ASK1 levels were regulated by cyclin D1, via the Rb-E2F pathway. Exogenous ASK1 induced cyclin D1 expression, followed by elevated expression of endogenous ASK1. These results indicate an autoregulatory mechanism of ASK1 in the development of gastric cancer. Targeting this positive feedback loop, ASK1 may present a potential therapeutic target for the treatment of advanced gastric cancer.

Extracellular vesicles in fatty liver disease and steatohepatitis: Role as biomarkers and therapeutic targets

Summary: Non-alcoholic fatty liver disease (NAFLD) and alcohol-associated liver disease (ALD) both involve lipid deposition in hepatocytes, with NAFLD occurring without excessive alcohol consumption and ALD occurring with excessive alcohol consumption. These conditions range from simple steatosis to more severe forms of liver disease. The term metabolic associated fatty liver disease (MAFLD) has been proposed to better reflect the risk factors and progression of the disease. Extracellular vesicles (EVs) released by cells contain cellular cargo and can serve as biomarkers for identifying the degree and type of liver disease. Mesenchymal stem cell (MSC)-derived EVs (MSC-EVs) have shown promise in regulating the immune response and are being investigated as a treatment for liver disease.

Proton beam therapy versus radiofrequency ablation for patients with treatment-naive single hepatocellular carcinoma: a propensity score analysis

Summary: This study compared the survival outcomes of proton beam therapy (PBT) with radiofrequency ablation (RFA) in treatment-naive solitary hepatocellular carcinoma (HCC). The results showed that after propensity score matching, there was no significant difference in recurrence-free survival (RFS) and overall survival (OS) between PBT and RFA. PBT can be considered as an alternative treatment for patients with solitary treatment-naive HCC.

Methylated SEPT9 assay-based liquid biopsy as a biomarker in molecular targeted agent-treated hepatocellular carcinoma

Summary: The potential of methylated SEPT9 gene as a biomarker for the treatment efficacy of molecular targeted agents (MTAs) in hepatocellular carcinoma (HCC) was investigated. The study analyzed 157 patients receiving sorafenib or lenvatinib as a first-line therapy and found that AFP level, methylated SEPT9 (m-SEPT9) level, and Eastern Cooperative Oncology Group (ECOG) performance status were significant predictors of overall survival (OS) in HCC patients. A predictive score called the MTA score was developed based on these predictors, and it showed that patients with MTA scores >= 2 had a significantly poor prognosis compared to those with MTA scores >= 1.

Steroid administration for ischemic complications after radiofrequency ablation: A retrospective study

Summary: The aim of this study was to evaluate the effects of steroids on ischemic complications after radiofrequency ablation. A total of 58 patients with ischemic complications were divided into two groups according to corticosteroid use or non-use. The results showed that patients who were administered steroids had a shorter duration of fever than those who were not administered steroids, and steroid administration was associated with a reduction in the duration of fever.

This suggests that Pemafibrate may be a promising therapeutic agent for NAFLD patients.

Summary: We report a case of steatohepatitis that occurred after pancreaticoduodenectomy (PD) and led to decompensated liver cirrhosis and hepatocellular carcinoma (HCC). Our findings indicate that progression to liver cirrhosis and HCC may become a severe problem in long-term post-PD survival, highlighting the need for further research on the mechanism of steatohepatitis after PD and the development of appropriate therapeutic strategies.
