Pituitary adenylyl cyclase-activating polypeptide ( PACAP) is a widely expressed neuropeptide originally discovered in the hypothalamus. It closely resembles vasoactive intestinal peptide ( VIP), a neuropeptide well known to inhibit macrophage activity, promote Th2-type responses, and enhance regulatory T cell (Treg) production. Recent studies have shown that administration of PACAP, like VIP, can attenuate dramatically the clinical and pathological features of murine models of autoimmune diseases such as experimental autoimmune encephalomyelitis (EAE) and collagen-induced arthritis. However, specific roles ( if any) of endogenous VIP and PACAP in the protection against autoimmune diseases have not been explored. Here, we subjected PACAP-deficient mice to myelin oligodendrocyte glycoprotein (MOG(35-55))-induced EAE. MOG immunization of PACAP-deficient mice triggered heightened clinical and pathological manifestations of EAE compared to wild-type mice. The increased sensitivity was accompanied by enhanced mRNA expression of proinflammatory cytokines (TNF alpha, IL-6, IFN-gamma, IL-12p35, IL-23p19, and IL-17), chemokines (MCP-1/CCL2, MIP-1 alpha/CCL3, and RANTES/CCL5), and chemotactic factor receptors (CCR1, CCR2, and CCR5), but downregulation of the anti-inflammatory cytokines (IL-4, IL-10, and TGF-beta) in the spinal cord. Moreover, the abundance of CD4(+)CD25(+)FoxP3(+) Tregs in lymph nodes and levels of FoxP3 mRNA in the spinal cord were also diminished. The reduction in Tregs was associated with increased proliferation and decreased TGF-beta secretion in lymph node cultures stimulated with MOG. These results demonstrate that endogenous PACAP provides protection in EAE and identify PACAP as an intrinsic regulator of Treg abundance after inflammation.

Summary: Incomplete pregnancies are associated with reduced risk of invasive epithelial ovarian cancer, with an inverse association observed for all major histotypes. A greater risk reduction was seen in clear cell ovarian cancer. Future research should focus on understanding the mechanisms underlying this reduced risk.

Summary: The use of combined oral contraceptives and progestin-only contraceptives is associated with a decreased risk of ovarian cancer. Specifically, the injectable progestin-only contraceptive depot-medroxyprogesterone acetate (DMPA) was found to be associated with a 35% decreased risk of ovarian cancer, with a significant trend of decreasing risk with increasing duration of use. Some studies showed an inverse association between DMPA use and ovarian cancer risk, while others showed increased risk, indicating a need for further investigation into the mechanism of this association.

Thyroid cancer incidence disparities among ethnic Asian American populations, 1990-2014

Summary: There are significant differences in thyroid cancer incidence and trends among disaggregated Asian and/or Pacific Islander ethnic subgroups in the United States. Filipinos have the highest incidence rate, while Japanese have the lowest. Asian Indians and/or Pakistanis show an increasing trend in thyroid cancer incidence among females, while Koreans show an increasing trend among males. Disaggregating APIs in research provides critical information for understanding thyroid cancer risk.

Summary: The study evaluated the association between hysterectomy and ovarian cancer, finding a complex relationship that is influenced by hormone therapy use and history of endometriosis. Adjustment for duration of hormone therapy use and endometriosis history is essential to fully understand the association between hysterectomy and ovarian cancer risk. The findings suggest that overall, hysterectomy is not a protective factor against ovarian cancer, but a potential inverse association exists for women with a history of endometriosis.

Association of environmental phenols with endometriosis and uterine leiomyoma: An analysis of NHANES, 2003-2006

Summary: This study examined the association between exposure to phenols and female reproductive health. The results showed a significant positive association between bisphenol A (BPA) exposure and endometriosis, but no significant association with fibroids. The study suggests that BPA may play a role in the pathogenesis of endometriosis.

Summary: This study analyzed the association between 12 lifestyle and personal exposures and having residual disease after surgery in patients with high-grade serous ovarian cancer (HGSC). The use of menopausal estrogen-only therapy (ET) was associated with a lower likelihood of having macroscopic residual disease, while parous women who did not breastfeed also had a lower likelihood of residual disease. These factors could potentially be included in risk stratification models for HGSC patients.
