IL-33, a new member of the IL-1 family, signals through its receptor ST2 and induces T helper 2 (Th2) cytokine synthesis and mediates inflammatory response. We have investigated the role of IL-33 in antigen-induced hypernociception. Recombinant IL-33 induced cutaneous and articular mechanical hype rn ociception in a time- and dose-dependent manner. The hypernociception was inhibited by soluble (s) ST2 (a decoy receptor of IL-33), IL-1 receptor antagonist (IL-1ra), bosentan [a dual endothelin (ET)(A)/ETB receptor antagonist], clazosentan (an ETA receptor antagonist), or indomethacin (a cyclooxygenase inhibitor). IL-33 induced hypernociception in IL-18(-/-) mice but not in TNFR1(-/-) or IFN gamma(-/-) mice. The IL-33-induced hypernociception was not affected by blocking IL-15 or sympathetic amines (guanethidine). Furthermore, methylated BSA (mBSA)-induced cutaneous and articular mechanical hypernociception depended on TNFR1 and IFN gamma and was blocked by sST2, IL-1ra, bosentan, clazosentan, and indomethacin. mBSA also induced significant IL-33 and ST2 mRNA expression. Importantly, we showed that mBSA induced hypernociception via the IL-33 -> TNF alpha -> IL-1 beta -> IFN gamma -> ET-1 -> PGE(2) signaling cascade. These results therefore demonstrate that IL-33 is a key mediator of immune inflammatory hype rn ociception normally associated with a Th1 type of response, revealing a hitherto unrecognized function of IL-33 in a key immune pharmacological pathway that may be amenable to therapeutic intervention.

Tumour necrosis factor plays a deleterious role in the pathogenesis of chikungunya virus infection

Thaiane Pinto Moreira, Carla Daiane Ferreira de Sousa, Victor Rodrigues de Melo Costa, Celso Martins Queiroz-Junior, Franciele Martins Santos, Caio Santos Bonilha, Lisia Maria Esper, Mauricio Lacerda Nogueira, Thiago Mattar Cunha, Mauro Martins Teixeira, Vivian Vasconcelos Costa, Daniele da Gloria de Souza

Summary: This study describes a novel mouse model of CHIKV infection and evaluates the role of tumour necrosis factor in the disease's pathogenesis. It shows that TNF plays a dual role in viral clearance and driving tissue damage and hypernociception. Therefore, TNF inhibitors may have therapeutic benefits, but viral levels must be monitored and used in combination with antiviral drugs when treating CHIKV-infected patients.

Summary: This study evaluated whether colchicine treatment was associated with the inhibition of NLRP3 inflammasome activation in patients with COVID-19. The results showed that colchicine reduced the need for supplemental oxygen and the length of hospitalization, and lowered the serum levels of Casp1p20 and IL-18 in the first two days of treatment.

HBP/O-GlcNAcylation Metabolic Axis Regulates Bone Resorption Outcome

T. M. Taira, E. S. Ramos-Junior, P. H. Melo, C. C. Costa-Silva, M. G. Alteen, D. J. Vocadlo, W. B. Dias, F. Q. Cunha, J. C. Alves-Filho, K. Soe, S. Y. Fukada

Summary: Osteoclasts are important in bone mass regulation and have high metabolic activity. The study shows that metabolic reprogramming towards the hexosamine biosynthetic pathway (HBP) is necessary for osteoclast differentiation and determining bone resorption modes. In addition, O-GlcNAcylation facilitates aggressive trench resorption in human cells.

Cunha, Alexandre T. Fabro, Paulo Louzada-Junior, Rene D. R. de Oliveira, Dario S. Zamboni

Summary: SARS-CoV-2 infection activates the NLRP3 inflammasome and leads to inflammation and severe COVID-19. Human caspase-4 (mouse caspase-11) is upregulated and activated by SARS-CoV-2, contributing to NLRP3 activation. In vivo studies using transgenic hACE2 humanized mice show that hACE2 Casp11(-/-) mice are protected from disease development, with reduced lung damage, clinical symptoms, and mortality. Analysis of human samples from fatal COVID-19 cases reveals the expression of CASP4 in patient lungs, which correlates with inflammasome components and inflammatory mediators. These findings suggest CASP4/11 as a potential target for therapeutic interventions in COVID-19.

Kaurenoic Acid Reduces Ongoing Chronic Constriction Injury-Induced Neuropathic Pain: Nitric Oxide Silencing of Dorsal Root Ganglia Neurons

Tiago H. Zaninelli, Sandra S. Mizokami, Mariana M. Bertozzi, Telma Saraiva-Santos, Felipe A. Pinho-Ribeiro, Gabriele Inacio de Oliveira, Renata Streck, Eduardo J. A. Araujo, Nilton S. Arakawa, Sergio M. Borghi, Rubia Casagrande, Waldiceu A. Verri

Summary: In this study, it was found that kaurenoic acid (KA), extracted from Sphagneticola trilobata, has analgesic properties and can be used for the treatment of neuropathic pain. KA inhibits neuropathic pain induced by chronic constriction injury (CCI) by activating the NO/cGMP/PKG/ATP-sensitive potassium channel signaling pathway. KA also reduces activation of sensory neurons and increases the levels of neuronal nitric oxide synthase (nNOS) and nitric oxide (NO) in dorsal root ganglion (DRG) neurons. These findings suggest that KA inhibits CCI neuropathic pain by activating the nNOS-NO pathway to silence nociceptive signaling.

Summary: The aim of this study was to evaluate oxidative stress in ovaries and corpus luteum (CL) of Bos taurus indicus females and the oxidant effect of CL in ovarian tissues. The results showed that luteal tissues had higher antioxidant activity than ovarian tissues, and there was no difference in antioxidant activity and oxidative stress between ovarian fragments obtained from different regions (with or without CL). The presence of CL in the ovaries did not influence oxidative stress or antioxidant potential.

Pre-emergence herbicidal efficiency and uptake of atrazine-loaded zein nanoparticles: a sustainable alternative to weed control

Lucas Braganca Carvalho, Isabela Silva Godoy, Ana Cristina Preisler, Patricia Luiza de Freitas Proenca, Telma Saraiva-Santos, Waldiceu Aparecido Verri Jr, Halley Caixeta Oliveira, Giliardi Dalazen, Leonardo Fernandes Fraceto

Summary: In this study, zein nanoparticles associated with the herbicide atrazine were developed and their efficacy in weed control was evaluated. The nanoformulation showed good stability and encapsulation efficiency. It exhibited greater effectiveness against the target plant, Brassica juncea, even at low doses, without causing toxicity to the crop species. The nanoparticles were captured and accumulated in roots but had limited transportation to the shoot. The atrazine nanoencapsulation did not increase the mobility of the herbicide in the soil.
