Modulation of the glutamatergic transmission by Dopamine: a focus on Parkinson, Huntington and Addiction diseases

Dopamine (DA) plays a major role in motor and cognitive functions as well as in reward processing by regulating glutamatergic inputs. In particular in the striatum the release of DA rapidly influences synaptic transmission modulating both AMPA and NMDA receptors. Several neurodegenerative and neuropsychiatric disorders, including Parkinson, Huntington and addiction related diseases, manifest a dysregulation of glutamate and DA signaling. Here, we will focus our attention on the mechanisms underlying the modulation of the glutamatergic transmission by DA in stnatal circuits.

Summary: Autoimmunity plays an expanding role in CNS disorders, with different types of autoantibodies targeting ionotropic glutamate receptors identified in patients with brain disorders. Research has improved our understanding of the effects induced by NMDAR and AMPAR autoantibodies at excitatory glutamatergic synapses. Animal studies have shown that autoantibodies can modulate the localization of target subunits at synapses, leading to functional impairments and behavioral alterations.

Summary: Mutations in the SHANK3 gene have been identified as a genetic risk factor for Autism Spectrum Disorder (ASD). This study demonstrates that region-specific downregulation of Shank3 in neonatal mice leads to hyperexcitability of certain neurons and impairs social behavior. Furthermore, acute inhibition of Trpv4 can rescue these behavioral deficits.

Summary: A prominent feature of neurodegenerative diseases is synaptic dysfunction and spine loss. Misfolded proteins, such as tau and alpha-synuclein, play a critical role in driving synaptic toxicity at excitatory glutamatergic synapses. Understanding the role of these proteins in impairing the function of glutamate receptors can provide insights into the pathology of neurodegenerative diseases.

Shank3 deficits in the anteromedial bed nucleus of the stria terminalis trigger an anxiety phenotype in mice

Summary: Anxiety disorders are the most common co-morbidity factor associated with autism spectrum disorders (ASD). The role of Shank3 protein in anxiety behaviors and the neuronal mechanisms underlying the co-occurrence of ASD and anxiety disorders are still poorly understood. In this study, the researchers characterized the developmental trajectories of locomotor, social behavior, and anxiety traits in a mouse model of ASD. They found that Shank3 downregulation in the anteromedial bed nucleus of the stria terminalis (amBNST) induced anxiety and social avoidance behaviors, accompanied by alterations in glutamatergic synaptic functions. These findings highlight the important role of Shank3 in the maturation of amBNST and anxiety control.

Intensive exercise ameliorates motor and cognitive symptoms in experimental Parkinson's disease restoring striatal synaptic plasticity

Summary: Intensive physical activity improves motor functions and cognitive deficits in early-stage Parkinson's disease (PD) patients. This study investigated the mechanisms behind these beneficial effects by conducting an intensive treadmill training program on rats with alpha-synuclein (a-syn) preformed fibrils. The training resulted in improved motor control, visuospatial learning, and plasticity in the brain, which were associated with the modulation of specific receptors. These findings suggest that intensive exercise can rescue early neuroplastic alterations caused by a-syn aggregates and reduce their spread to vulnerable brain areas.
