Phylogenetic diversity of Gram-positive bacteria cultured from Antarctic deep-sea sponges

Gram-positive bacteria, specifically actinobacteria and members of the order Bacillales, are well-known producers of important secondary metabolites. Little is known about the diversity of Gram-positive bacteria associated with Antarctic deep-sea sponges. In this study, cultivation-based approaches were applied to investigate the Gram-positive bacteria associated with the Antarctic sponges Rossella nuda, Rossella racovitzae (Porifera: Hexactinellida), and Myxilla mollis, Homaxinella balfourensis, Radiella antarctica (Porifera: Demospongiae). In total, 46 Gram-positive strains were cultured. Phylogenetic analysis revealed that 24 strains were affiliated with the Actinobacteria, including six genera Streptomyces, Nocardiopsis, Pseudonocardia, Dietzia, Brachybacterium, and Brevibacterium. The other 22 strains were affiliated with the Firmicutes, and among them two (V17-1 and V179-1) only shared 92-95% 16S rRNA gene sequence identity with the nearest type strain. To our knowledge, this is the first report on the isolation of strains belonging to genera Dietzia and Brevibacterium from Antarctic sponges. All of the 46 strains were PCR screened for genes encoding polyketide synthases (PKS), and a selection of 36 isolates were used in subsequent bioassay analyses. Eighty-eight percentage of the isolates that possess a PKS gene were active against at least one test organism. The study confirms the existence of diverse bacteria in Antarctic sponges and their potential for producing active compounds.

Summary: This study reports a fast and sustainable technique for catalytically degrading organic dyes under flow. By utilizing a vortex fluidic device (VFD) with different applications, the researchers have successfully fabricated a composite catalytic material with enhanced activity. Furthermore, chemical modification of the VFD tube reactor has dramatically improved the degradation efficiency. Real-time monitoring of the reaction and evaluation of the coating layer post-reaction were achieved using a UV-visible spectroscopy-based probe.

Astragalus Shiitake-A Novel Functional Food with High Polysaccharide Content and Anti-Proliferative Activity in a Colorectal Carcinoma Cell Line

Summary: This study compared the yield and antiproliferative activity of polysaccharide-rich extracts from Hengshan Astragalus Shiitake (HAS) using three different extraction techniques. The results showed that both HAS-A and HAS-B extracts contained significantly higher amounts of polysaccharides compared to the control (Shiitake extract). The HAS-B extract also exhibited significantly higher antiproliferative activity in human colorectal carcinoma cells when using the ultrasound-enzyme assisted extraction method.

Marine Sponge Endosymbionts: Structural and Functional Specificity of the Microbiome within Euryspongia arenaria Cells

Summary: Sponges host abundant microbial symbionts that can produce an impressive number of novel bioactive metabolites. However, knowledge on intracellular (endosymbiotic) microbiota is scarce. By separating sponge cells and enriching intracellular microbes, we characterize the composition and function of the endosymbiotic microbiome, uncovering a noteworthy number of taxa exclusively in the endosymbiotic microbiome. Our study achieves a more comprehensive sponge microbial community profile, which demonstrates the structural and functional specificity of the endosymbiotic microbiome.

Bioactives From Marine Animals: Potential Benefits for Human Reproductive Health

Summary: This review assesses the potential of marine-derived bioactives in improving reproductive and sexual health issues. While there have been some advancements, there are still gaps in knowledge and technology that need further research. Developing simple, low-cost, and scalable processes for extraction and purification, as well as optimizing culturing conditions for sustainable production, are suggested. Mechanistic studies and clinical trials are urgently needed to evaluate the efficacy and safety of extracts and marine-derived bioactives.

Printable Hydrogel Arrays for Portable and High-Throughput Shear-Mediated Assays

Summary: A shear-stress-mediated platform called portable vortex-fluidic device (P-VFD) has been developed to overcome the diffusion-limit and achieve fast reaction rates. The platform consists of a plasma oxazoline-coated polyvinyl chloride (POx-PVC) film with tough hydrogel covalently bound to its surface and a reactor tube for reactions. The hydrogel arrays on the film provide a strong matrix for entrapping biomolecules and tolerate shear stress, resulting in a >6-fold increase in reaction rate.

Magnetic Fe3O4@ZIF-8 nanoparticles as a drug release vehicle: pH-sensitive release of norfloxacin and its antibacterial activity

Summary: Metal-organic frameworks (MOFs) with high surface area and biodegradability have potential in drug release. Fe3O4 nanoparticles and MOFs composite materials, with magnetic features, were investigated for norfloxacin loading and release. The composite showed pH-sensitive properties and high release efficiency, making it suitable for targeted drug delivery. Its antibacterial activity against Escherichia coli was also tested, indicating its potential as a microbial inhibitor.

Summary: A ligand-mediated, spatially-confined synthesis method is developed to produce high-quality carbon dots from crude bioresources. The choice of solvent plays a critical role in the formation and properties of the carbon dots. The ligand-carbon interactions and electron transfer result in the formation of uniform colloidal carbon dots with multiple emissions. These carbon dots can be easily dispersed in various solvents and fabricated into thin-film devices, making them suitable for optoelectronic applications.

Anti-skin glycation and collagen level stimulation of brown seaweed extracts and their compositional characteristics

Summary: This study investigated the anti-skin aging potential of three Southern Australian brown seaweed extracts. The results showed that the extracts from Cystophora moniliformis and Cystophora siliquosa exhibited strong inhibitory effects on glycation activity and stimulated collagen production, suggesting their potential as skin anti-aging supplements. Further in vivo studies are warranted.
