Repurposing antimalarial aminoquinolines and related compounds for treatment of retinal neovascularization

Neovascularization is the pathological driver of blinding eye diseases such as retinopathy of prematurity, proliferative diabetic retinopathy, and wet age-related macular degeneration. The loss of vision resulting from these diseases significantly impacts the productivity and quality of life of patients, and represents a substantial burden on the health care system. Current standard of care includes biologics that target vascular endothelial growth factor (VEGF), a key mediator of neovascularization. While anti-VGEF therapies have been successful, up to 30% of patients are non-responsive. Therefore, there is a need for new therapeutic targets, and small molecule inhibitors of angiogenesis to complement existing treatments. Apelin and its receptor have recently been shown to play a key role in both developmental and pathological angiogenesis in the eye. Through a cell-based high-throughput screen, we identified 4-aminoquinoline antimalarial drugs as potent selective antagonists of APJ. The prototypical 4-aminoquinoline, amodiaquine was found to be a selective, non-competitive APJ antagonist that inhibited apelin signaling in a concentration-dependent manner. Additionally, amodiaquine suppressed both apelin-and VGEF-induced endothelial tube formation. Intravitreal amodaiquine significantly reduced choroidal neovascularization (CNV) lesion volume in the laser-induced CNV mouse model, and showed no signs of ocular toxicity at the highest doses tested. This work firmly establishes APJ as a novel, chemically tractable therapeutic target for the treatment of ocular neovascularization, and that amodiaquine is a potential candidate for repurposing and further toxicological, and pharmacokinetic evaluation in the clinic.

Faster CNN-based vehicle detection and counting strategy for fixed camera scenes

Summary: This paper proposes an efficient real-time approach for the detection and counting of moving vehicles based on YOLOv2 and features point motion analysis. By synchronously detecting and tracking vehicle features, accurate counting results are achieved. Experimental results show that the proposed method outperforms existing strategies and improves computational efficiency.

Summary: Esophageal adenocarcinoma cells exploit the unfolded protein response for survival, with Aurora kinase A hijacking the pro-survival unfolded protein response in promoting cancer cell survival under stressful conditions. Targeting Aurora kinase A can reverse prosurvival unfolded protein response mechanisms and reduce cancer cell survival, offering a promising treatment approach for esophageal adenocarcinoma patients.

A wide axial-ratio beamwidth circularly-polarized oval patch antenna with sunlight-shaped slots for gnss and wimax applications

Summary: This paper proposes a quadruple band stacked oval patch antenna that supports L1/L2/L5 GNSS bands and the 2.3 GHz WiMAX band. By etching sunlight-shaped slots into the patches, the antenna is able to fine-tune the phase shifts between different modes versions, resulting in enhanced axial-ratio beamwidth. Despite its simple structure and feeding mechanism, the proposed antenna achieves wider axial-ratio beamwidths compared to previous works.

Seismic hazard evaluation by employing microtremor measurements for Abu Simbel area, Aswan, Egypt

Summary: The Abu Simbel area in Egypt contains important temples, and the use of microtremor measurement has allowed for evaluation of the dynamic properties and vulnerability of the temples and surrounding region. The seismic vulnerability index was found to be consistent with the amplification factor, indicating the potential for earthquake disasters.

This study provides cryoEM structures of NTSR1 complexes with Gq and GoA, and discusses the allosteric modulation of NTSR1 by the small-molecule SBI-553.

This study focuses on developing a digital documentation process for Abu Simbel Temple in Egypt using laser scanning technology. The result is a 3-D digital model that includes various data and a virtual tour, which can help raise heritage awareness, promote tourism, and aid in future restoration efforts.
