Resveratrol downregulates inflammatory pathway activated by lymphotoxin α (TNF-β) in articular chondrocytes: Comparison with TNF-α

While Lymphotoxin a (TNF-beta), a product of lymphocytes, is known to play a pivotal role in inflammatory joint environment, resveratrol has been shown to possess anti-inflammatory and chondroprotective effects via activation of the histondeacetylase Sirt1. Whether TNF-beta induction of inflammatory pathways in primary human chondrocytes (PCH) can be modulated by resveratrol, was investigated. Monolayer and alginate cultures of PCH were treated with TNF-beta, anti-TNF-beta, nicotinamide (NAM), antisense oligonucleotides against Sirt1 (Sirt1-ASO) and/or resveratrol and co-cultured with T-lymphocytes. We found that resveratrol suppressed, similar to anti-TNF-beta, TNF-beta-induced increased adhesiveness in an inflammatory microenvironment of T-lymphocytes and PCH. In contrast, knockdown of Sirt1 by mRNA abolished the inhibitory effects of resveratrol on the TNF-beta-induced adhesiveness, suggesting the essential role of this enzyme for resveratrol-mediated anti-inflammatory signaling. Similar results were obtained in PCH stimulated with TNF-alpha. Sirt1-ASO, NAM or TNF-beta, similar to T-lymphocytes induced inflammatory microenvironment by down-regulation of cartilage-specific proteins, Sox9, Ki67 and enhanced NF-kappa B-regulated gene products involved in inflammatory and degradative processes in cartilage (MMP-9/-13, COX-2, caspase-3), NF-kappa B activation and its translocation to the nucleus. Moreover, resveratrol reversed the TNF-beta-, NAM-, T-lymphocytes-induced up-regulation of various NF-kappa B-regulated gene products. Down-regulation of Sirt1 by mRNA interference abrogated the effect of resveratrol on TNF-beta-induced effects. Ultrastructural and cell viability assay investigations revealed that resveratrol revoked TNF-beta-induced dose-dependent degradative/apoptotic morphological changes, cell viability and proliferation in PCH. Taken together, suppression of TNF-beta-induced inflammatory microenvironment in PCH by resveratrol/Sirt1 might be a novel therapeutic approach for targeting inflammation during rheumatoid arthritis.

Evidence That beta 1-Integrin Is Required for the Anti-Viability and Anti-Proliferative Effect of Resveratrol in CRC Cells

Summary: This study found that resveratrol can exert its suppressive actions on tumor microenvironment (TME)-induced tumorigenesis in colorectal cancer (CRC) cells through the beta 1-integrin receptor. Resveratrol altered the distribution pattern and expression of beta 1-integrin on the surface of CRC cells in TME, leading to decreased cell proliferation and viability, and increased apoptosis. Inhibition of beta 1-integrin or interruption of downstream signaling pathway attenuated the anti-tumor effects of resveratrol. The results highlight the important role of beta 1-integrins as signal carriers for resveratrol in CRC cells.

Probiotics, prebiotics, and synbiotics in type 1 diabetes mellitus: A systematic review and meta-analysis of clinical trials

Summary: Dysbiosis in the gut has been linked to T1DM, and this review analyzed the effects of probiotics, prebiotics, and synbiotics on T1DM patients. After evaluating 258 entries, five trials were included, showing that probiotic supplementation decreased fasting blood glucose but had no significant effect on HbA1c, C-peptide, and insulin requirements. Probiotics may serve as a complementary therapeutic strategy for T1DM, but more trials are needed.

Curcumin, calebin A and chemosensitization: How are they linked to colorectal cancer?

Summary: Colorectal cancer (CRC) is a major global malignant disease often resistant to chemotherapy drugs. Two polyphenolic turmeric ingredients, calebin A and curcumin, show anti-inflammatory and anticancer effects, including the ability to sensitize CRC cells to chemotherapy drugs. They modulate inflammation, proliferation, cell cycle, cancer stem cells, and apoptotic signaling to convert chemoresistant CRC cells into non-chemoresistant cells. Further studies and clinical trials are needed to test their effectiveness in overcoming cancer chemoresistance.
