Summary: In this study, a gene-edited cashmere goat with improved fiber length and yield was generated by integrating the VEGF gene at the FGF5 site. The up-regulation of VEGF and down-regulation of FGF5 affected cell cycle, proliferation, and vascular tone through the PI3K-AKT signaling pathway and extracellular matrix-receptor interactions, resulting in impressive cashmere performance of the gene-edited cashmere goat.

Summary: This study investigated the use of CRISPR/Cas9 technology for transgenic improvement of channel catfish, showing that knock-in technology can increase the content of n-3 fatty acids. The results indicate that CRISPR/Cas9 technology can more effectively integrate transgenes and enhance the biological effects of foreign genes in fish.

Summary: CRISPR/Cas9-mediated knock-in is widely used in gene therapy, gene function study, and transgenic breeding programs. Researchers have developed a new strategy to improve knock-in efficiency and achieve multiple gene knock-ins simultaneously by targeting long repeated sequences in the genome.

Summary: Fluorescent reporter mouse lines and Cre/Flp recombinase driver lines are essential for investigating molecular functions in vivo. The study reverified and optimized the phospho-PCR method to obtain highly pure long ssDNAs for knock-in mouse generation, enabling successful generation of knock-in mice through microinjection and electroporation methods, thus broadening its applicability.

Summary: CRISPR-Cas9-based genome-editing is an efficient and cost-effective method to generate zebrafish loss-of-function alleles, but introducing patient-specific variants into the zebrafish genome remains challenging. This study illustrates an efficient approach to develop knock-in zebrafish models using sgRNA-Cas9 RNP complexes and single-stranded DNA templates, with improved knock-in efficiency achieved by morpholino-induced blocking of the DNA repair pathway.

Summary: Salicylic acid (SA) is an essential plant hormone that plays critical roles in plant defense. Three OsS5H homologues were found to convert SA into 2,5-dihydroxybenzoic acid (2,5-DHBA) in rice. Overexpression of OsS5H genes increased susceptibility to bacterial blight and rice blast. Triple mutants of oss5h1oss5h2oss5h3 displayed enhanced resistance to Xanthomonas oryzae pv. oryzae (Xoo) and rice blast.

Summary: Gene vectors deliver genetic materials or gene editing devices into cells for therapeutic protein production or gene correction, but barriers like cell membrane and degradation necessitate carriers for clinical gene therapy. Exosomes, engineered to encapsulate and deliver nucleic acids, and their fusion with liposomes increase loading capacity and maintain targeting capability, showing promise for future gene delivery applications in therapy.

Summary: In this study, various gene targeting strategies were compared to generate a novel knock-in reporter rat line. By visualizing the expression pattern of the transcription factor Tfap2c, this rat line can be used to identify and purify cells expressing Tfap2c in rats, making it a valuable resource.

Summary: By using CRISPR/Cas9 technology to insert the Akaluc gene at the AAVS1 locus, a clonal human embryonic stem stable cell line called AkalucHES was generated, which efficiently expresses Akaluc and can be easily traced in vivo, maintaining normal karyotype and capable of differentiating toward three germ layers.

Summary: This study investigated the use of two small molecules, B02 and Nocodazole, to improve CRIS-PITCh knock-in efficiency. The results showed that Nocodazole had a more significant effect than B02. The findings of this study could be useful for future research in establishing rCHO cell lines.

Summary: In this study, the role of BmMETTL4 in silkworm embryonic development was investigated. Mutations in BmMETTL4 resulted in developmental defects and lethality in late silkworm embryos. Analysis showed that BmMETTL4 mutation significantly affected genes involved in molecular structure, chitin binding, and serine hydrolase activity. Additionally, expression of cuticular protein genes and collagens was decreased while collagenases were increased, contributing to abnormal embryo development and decreased hatchability in silkworm.

Summary: Comprehensive and accurate analysis of respiratory and metabolic data is crucial for studying diseases, and the lack of tools for high-throughput analysis is a challenge. This study introduces a new open-source pipeline called Breathe Easy, which improves the efficiency of respiratory data analysis and generates effective results and publication-worthy statistical analysis graphs. The validity of this pipeline is confirmed through a long-term study on an Alzheimer's disease mouse model.
